Reactive Oxygen Species in Cancer Stem Cells

Abstract: Significance: Reactive oxygen species (ROS), byproducts of aerobic metabolism, are increased in many types of cancer cells. Increased endogenous ROS lead to adaptive changes and may play pivotal roles in tumorigenesis, metastasis, and resistance to radiation and chemotherapy. In contrast, the ROS generated by xenobiotics disturb the redox balance and may selectively kill cancer cells but spare normal cells. Recent Advances: Cancer stem cells (CSCs) are integral parts of pathophysiological mechanisms of tumor p… Show more

“…71 Nicotinamide's ability to overcome pluripotency deficits and reprogramming barriers has yet to be evaluated in the acquisition and maintenance of CSC cellular states and future studies should establish whether an adequate NAD C content is important for enhancing the resistance to stress in CSC. Accumulating evidence suggests that, in contrast to differentiated cancer cells where ROS levels are increased, CSC maintain low levels of ROS, [72][73][74][75] and thus exhibit redox patterns similar to the corresponding normal stem cell. Accordingly, an increased reliance on glucose metabolism reduces the levels of ROS to promote EMT and CSC-like phenotypes.…”

“…Apelin can inhibit the generation of ROS, which suggests that apelin suppresses tumorigenesis by regulating oxidative stress. However, studies have found that cancer stem cells, as a type of cell with the characteristics of stem cells in tumor tissues, can resist the inhibitory effects of oxidative stress caused by radiotherapy or chemotherapy (69)(70)(71). There is evidence that compared with ordinary tumor cells, there are fewer oxidation products of DNA, stronger resistance to oxidative stress, and higher activation level of the antioxidant stress system, existing in cancer stem cells (71).…”
