Real-time electrochemical detection of extracellular nitric oxide in tobacco cells exposed to cryptogein, an elicitor of defence responses

Abstract: It was previously reported that cryptogein, an elicitor of defence responses, induces an intracellular production of nitric oxide (NO) in tobacco. Here, the possibility was explored that cryptogein might also trigger an increase of NO extracellular content through two distinct approaches, an indirect method using the NO probe 4,5-diaminofluorescein (DAF-2) and an electrochemical method involving a chemically modified microelectrode probing free NO in biological media. While the chemical nature of DAF-2-reactiv… Show more

“…Therefore, the regulation of the expression of NAS4 might differ from those of IRT1 and FRO2. release of NO from DEANO occurs within seconds in aqueous solution 17 whereas the release form GSNO is slower. 18 Although speculative, we assume that these parameters might influence the action of NO.…”

“…GSNO and GSH were dissolved in water, DEANO was prepared as previously described. 17 As a control for DEANO effects, plants were treated with 50 μM diethylamine prepared in water.…”

“…The effect of salt was studied by complementing the hydroponic solution with 100 mM NaCl over 45 min or 2 h. For mannitol treatment, plants were grown in a hydroponic solution containing 200 mM mannitol over 45 min to 6 h. Plants were treated with 500 M H 2 O 2 over 8 or 20 min. Nitric oxide (NO) treatment was applied by using diethylamine NONOate (DEA-NO), an NO donor, as previously described (24). Briefly, DEA-NO was dissolved in 0.01 M NaOH as a stock solution (500 mM).…”

“…S1B). Pre-treatment of the root system with an NO scavenger (100 M cPTIO) (24,25) fully counteracted the inhibitory effects of 100 M DEA-NO (supplemental Fig. S1C), indicating that NO specifically inhibits Arabidopsis Lp r .…”

“…Peptidoglycan, a MAMP associated with gram-positive bacteria, also induces plant immune signaling dependent on [Ca 2+ ] cyt elevation and NO generation (Gust et al, 2007). The fungal MAMPs xylanase and chitosan also cause NO generation in plants (Laxalt et al, 2007;Raho et al, 2011), as do the Phytophthora infestans oomycete elicitins (INF1; Asai et al, 2008) and cryptogein (Foissner et al, 2000;Lamotte et al, 2004;Besson-Bard et al, 2008b). One of the best-characterized MAMPs, in terms of plant immune responses, is the bacterial motility organ protein flagellin (flg).…”
