Real‐Time Intracellular Measurements of ROS and RNS in Living Cells with Single Core–Shell Nanowire Electrodes

Abstract: Nanoelectrodes allow precise and quantitative measurements of important biological processes at the single living-cell level in real time. Cylindrical nanowire electrodes (NWEs) required for intracellular measurements create a great challenge for achieving excellent electrochemical and mechanical performances. Herein, we present a facile and robust solution to this problem based on a unique SiC-core-shell design to produce cylindrical NWEs with superior mechanical toughness provided by the SiC nano-core and an… Show more

“…[8] We recently reported the fabrication of anovel kind of platinized nanowire SiC@C electrodes (Pt-NWEs) and demonstrated their perfect suitability to analyze ROS/RNS inside single phagolysosomes of living macrophages. [9] Phagolysosomes may then randomly collide onto the surface of aP t-NWE inserted into am acrophage cytoplasm ( Figure 1A). This situation is identical to that which operates during vesicle impact electrochemical cytometry (VIEC), am ethod initially introduced for quantifying neurotransmitters amounts in liposomes and exocytotic vesicles.…”

“…Overactivation of glutamate receptors causes excitotoxicity, a pathological process whereby neurons are damaged or killed, which may result in neurodegeneration. 1,2 Because of its central role in metabolic and cognitive processes, a strong effort has been put toward developing methods to detect glutamate. [2][3][4][5] Accurate detection of glutamate can be accomplished using many techniques, including spectrometry, spectroscopy, and electrochemistry.…”

“…Nanoelectrochemical monitoring of intracellular ROS/RNS at the single cell level is a very efficient and quantitative method that has been applied in cancer cells, macrophages and neurons . We recently reported the fabrication of a novel kind of platinized nanowire SiC@C electrodes (Pt‐NWEs) and demonstrated their perfect suitability to analyze ROS/RNS inside single phagolysosomes of living macrophages . Phagolysosomes may then randomly collide onto the surface of a Pt‐NWE inserted into a macrophage cytoplasm (Figure A).…”
