Recent Advances and Opportunities in Research on Lupus: Environmental Influences and Mechanisms of Disease

Abstract: ObjectivesIn this review we summarize research on mechanisms through which environmental agents may affect the pathogenesis of lupus, discuss three exposures that have been the focus of research in this area, and propose recommendations for new research initiatives.Data sources and synthesisWe examined studies pertaining to key mechanistic events and specific exposures. “…When macrophages internalize erionite and asbestos fibers, they tend to produce reactive oxygen species (ROS) (Carbone et al 2012; Hogg et al 1996), which contribute to carcinogenicity and may also play a role in the pathogenesis of autoimmune diseases. The oxygen radicals may alter self-antigens in a way that causes the immune system to lose self-tolerance and mount an attack on those antigens (Cooper et al 2008).
