Recent advances of cocktail chemotherapy by combination drug delivery systems

Abstract: Combination chemotherapy is widely exploited for enhanced cancer treatment in clinic. However, the traditional cocktail administration of combination regimens often suffers from varying pharmacokinetics among different drugs. The emergence of nanotechnology offers an unparalleled opportunity for developing advanced combination drug delivery strategies with the ability to encapsulate various drugs simultaneously and unify the pharmacokinetics of each drug. This review surveys the most recent advances in combina… Show more

“…Traditional combinations include methotrexate-based combinations (methotrexate plus cyclophosphamide/5-fluorouracyl (5-FU)), anthracycline-based combinations (Dox plus 5-FU) and taxane-based combinations (PTX plus cisplatin/carboplatin) [16]. Platinum compounds are also used in combination with other chemotherapeutic drugs in a clinical setting.…”

“…Chemotherapy is also combined with a photothermal agent, a compound that generates cell-damaging hyperthermia in response to light stimulation [16], such as indocyanine dye [81], gold nanorods [82], and graphene [83]. For example, PTX was combined with indocyanine green (ICG) with human serum albumin (HSA) as a common carrier [84].…”

“…This situation can be improved to some extent by cocktail or combination chemotherapy (i.e., the simultaneous administration of two or multiple therapeutic agents or drugs), of which the preliminary trials include doxorubicin (DOX), paclitaxel, and cisplatin based chemotherapeutics. [1][2][3] However, the traditional cocktail administration still suffers from uncontrollable pharmacokinetics among different drugs. On the other hand, the polymeric drug-loaded nanoparticles (i.e., the nanomedicine formulations) have presented distinct advantages including the enhancement of drug solubility, the prolongation of blood circulation time, the improvement of pharmacokinetics and pharmacodynamics, and especially the enhanced permeation and retention (EPR) effect in solid tumors compared to free drugs.…”

“…At last, the nucleus targeting anticancer drugs need to be further delivered to the nucleus for actions; the nuclear envelope is also the obstruction. In brief, we believe that continuous research efforts on NDCDS will provide some solutions to breaking the biological barrier to achieve the goal of effective cancer treatment (Hu et al, 2016).…”
