The use of the insect cell‐baculovirus system has grown significantly over the past three decades. The present chapter elucidates the fundamentals, scientific developments, technologies, and hurdles encountered in this field. The common types of insects and insect lines used as well as their specific features and culture conditions are described. Additionally, the types of baculoviruses used and the methods for generating them are also revealed. Particular emphasis has been given to describing the construction of recombinant baculoviruses that are used in production of heterologous proteins with insect cells. A major advantage in using insect cells is their ability to perform many post‐translational processing events on these proteins including glycosylation. The types of insect cell glycosylation and the differences in glycosylation patterns between insect and mammalian systems are detailed. The parameters that affect insect cell growth and baculovirus infection including bioreactor configuration, process monitoring and control, oxygen supply, sparging, and media formulations are described. In particular, cell culture processing methods including batch, fed‐batch, perfusion, and semi‐batch processes are illustrated. Additional factors affecting baculovirus infection including multiplicity, time of infection, and the generation of mutant viruses are also considered. A final section is devoted to the use of insect larvae in combination with baculovirus as a potentially alternative to cell culture for the low‐cost generation of baculoviruses and heterologous proteins. With all the advances in molecular biology and engineering processes for the baculovirus‐insect cell system, insect cell culture and its application to recombinant protein production, vaccines, pesticides, and other areas will continue to expand in the coming decades.

Three popular expression host systems Escherichia coli, Pichia pastoris and Drosophila S2 were analyzed techno-economically using HIV-1 Nef protein as the model product. On scale of 100 mg protein, the labor costs corresponded to 52-83% of the manufacturing costs. When analyzing the cost impact of the different phases (strain/cell line construction, bioreactor production, and primary purification), we found that with the microbial host systems the strain construction phase was most significant generating 56% (E. coli) and 72% (P. pastoris) of the manufacturing costs, whereas with the Drosophila S2 system the cell line construction and bioreactor production phases were equally significant (46 and 47% of the total costs, respectively). With different titers and production goal of 100 mg of Nef protein, the costs of P. pastoris and Drosophila S2 systems were about two and four times higher than the respective costs of the E. coli system. When equal titers and bioreactor working volumes (10 L) were assumed for all three systems, the manufacturing costs of the bioreactor production of the P. pastoris and Drosophila S2 systems were about two and 2.5 times higher than the respective costs of the E. coli system. We identified 1219 articles published in 2006 that described work performed using commercial optical biosensor platforms. It is interesting to witness how the biosensor market is maturing with an increased number of instrument manufacturers offering a wider variety of platforms. However, it is clear from a review of the results presented that the advances in technology are outpacing the skill level of the average biosensor user. While we can track a gradual improvement in the quality of the published work, we clearly have a long way to go before we capitalize on the full potential of biosensor technology. To illustrate what is right with the biosensor literature, we highlight the work of 10 groups who have their eye on the ball. To help out the rest of us who have the lights on but nobody home, we use the literature to address common myths about biosensor technology.
