Recombinant Toxoplasma gondii Ribosomal Protein P2 Modulates the Functions of Murine Macrophages In Vitro and Provides Immunity against Acute Toxoplasmosis In Vivo

Abstract: Almost every warm-blooded animal can be an intermediate host for Toxoplasma gondii (T. gondii); there is still no efficient vaccine and medicine available for T. gondii infections. Detected on the surface of free tachyzoites of T. gondii, T. gondii ribosomal protein P2 (TgRPP2) has been identified as a target for protection against toxoplasmosis. In the present study, TgRPP2 was firstly expressed in a prokaryotic expression system, and the purified recombinant TgRPP2 (rTgRPP2) was characterized by its modulati… Show more

“…The PCR product was purified by Gel-Extraction Kit (Omega, Biotech, Norcross, GA, USA) and ligated into a pMD19-T cloning vector (TaKaRa, Dalian, China). The recombinant (pMD19-T/ Ts-MAPRC2 ) plasmid was further processed into the E. coli (DH5α) strain (Invitrogen, Shanghai, China) and cultivated in LB (Luria Bertani) [ 25 ] medium with ampicillin (100 μg/mL). Positive bacterial (pMD19-T/ Ts-MAPRC2 ) clones were assured by digestion of restriction enzymes and confirmed through sequencing (Invitrogen, China).…”

“…The total soluble protein of T. spiralis (muscle larvae) parasites was collected using RIPA solution (Thermo Scientific, Waltham, MA, USA) and a proteinase inhibitor (Thermo Scientific, Waltham, MA, USA) for complete lysis. The solution, containing the total soluble protein of T. spiralis was collected by high-speed centrifugation at 12,000 r/min for 30 min at 4 °C, and the supernatant was determined by the bicinchoninic acid method (BCA) kit mentioned above [ 25 ]. Afterwards, the total soluble proteins of T. spiralis (muscle larvae) parasites were carried by SDS-PAGE (12%) and transferred to polyvinylidene difluoride (PVDF) membranes (Millipore, Bedford, MA, USA) as described above.…”

“…Moreover, the highly expressed ribosomal proteins may represent an interesting pool of potent targets for the innate immune system. For instance, ribosomal protein P2 has been previously identified as a target for protection against toxoplasmosis 98 .Based on previous reports of the importance of MIC1, MIC2 and GRA2 for in effective immune evasion or virulence 40,99,100 , T. gondii genes encoding secreted proteins in ETGN 2 likely belong to a network of genes with importance for these processes. Conclusively, while not providing causality or evidence for direct interactions, which is beyond the scope of this study, we consider our findings a valuable resource to investigate potential direct interactions between genes that correlate positively in expression in single cells for future functional studies on host-pathogen interactions.…”
