Reconstruction of the right ventricular outflow tract with a bovine jugular vein graft fixed with a naturally occurring crosslinking agent (genipin) in a canine model

Abstract: Although further studies are necessary, the genipin-fixed valved conduit appears to have great potential in helping mitigate the complications observed in the commercially available conduits.

“…Anti-inflammatory properties are added values during genipin-induced cross-linking, showing to be 10, 000 times less cytotoxic then glutaraldehide which may produce weakly clastogenic responces in CHO-K1 cells (Tsai et al, 2000;Sisson et al, 2009). Moreover, the minimal calcium content of genipin-fixed tissue was detected (Chang, 2001). Genipin is a natural product, being obtained from an iridoid glucoside, geniposide abundantly present in Genipa Americana and Gardenia jasminoides Ellis.…”

“…Genipin was used as a cross-linking agent to crosslink the amino acid groups of gelatin as shown in Fig. 3 (24). Various concentrations of genipin (i.e., 3, 4, 5, and 6% w/w based on the weight of gelatin powder) were added into gelatin solutions, and then, the solutions were stirred at room temperature for 1 h. These solutions were then poured into polypropylene (PP) mold and lyophilized to form scaffolds by freeze-drying method.…”

“…Although the mechanism is not fully understood [10], it is known that genipin reacts with molecules that contain primary amines causing spontaneous irreversible pH-controlled CXL (Fig. 1).…”
