Regenerative Potential of Blood-Derived Products in 3D Osteoarthritic Chondrocyte Culture System

Abstract: Intra-articular injection of different types of blood-derived products is gaining popularity and clinical importance in the treatment of degenerative cartilage disorders such as osteoarthritis. The regenerative potential of two types of platelet-rich plasma (PRP), prepared in the presence of EDTA (EPRP) and citrate (CPRP) and an alternative blood product-hyperacute serum (hypACT) was evaluated using a 3D osteoarthritic chondrocyte pellet model by assessing the metabolic cell activity, cartilage-related gene ex… Show more

“…The in vitro studies demonstrated that treatment with HS resulted in increased viability of osteoarthritic chondrocytes and MSCs, reduced expression of pro-inflammatory cytokines, increased expression of antiinflammatory cytokines, increased differentiation of MSCs towards the chondrogenic lineage, and increased expression of chondrogenesis-related genes [11][12][13][14][15][16][17]. In addition, the cell-free and anti-coagulant-free nature of HS led to no variability in the differentiation potential or gene expression of MSCs, unlike PRP.…”

“…Kuten-Pella et al assessed and compared the regenerative potential of HS, EPRP, and CPRP using an osteoarthritic chondrocyte 3D pellet model by evaluating the metabolic activity of cells and the expression of genes within the pellet [ 15 ]. No significant differences in metabolic activity were observed between the three groups.…”

“…Kuten-Pella et al, using a 3D osteoarthritic chondrocyte pellet model, evaluated the regenerative potential of two types of PRP, one prepared with EDTA and citrate and an alternative blood product, hyperacute serum (hypACT). They obtained the best results for hypACT and stated that the possible molecular mechanisms of blood-derived products should be investigated in order to obtain the best results [52].…”
