Registration of Isogenic HMW-GS Ax1 Protein Deficient Lines of an Elite Wheat Cultivar

Abstract

A series of isogenic wheat (Triticum aestivum L.) lines deficient in high‐molecular weight subunit (HMW‐GS) proteins encoded on the A‐genome of an elite cultivar of bread wheat was developed by USDA–ARS at the Western Regional Research Center in Albany, CA: Ax1‐E28H (Reg. No. GS‐179, PI 674178); Ax1‐F259C (Reg. No. GS‐180, PI 674179); Ax1‐F261B (Reg. No. GS‐181, PI 674180); Ax1‐F80B (Reg. No. GS‐182, PI 674181); Ax1‐F83B (Reg. No. GS‐183, PI 674182). This set of A‐genome encoded HMW‐GS deficient lines was identified from a chemical‐ and radiation‐mutagenized population of hexaploid wheat cultivar Summit by screening M3 seeds for altered storage protein profiles using one‐dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS‐PAGE). The mutant lines are distinct from the wild‐type progenitor by the absence of the HMW‐GS Ax1 protein. Dough derived from greenhouse‐ and field‐grown plants of one mutant exhibited weaker dough but achieved shorter development time compared with the control. Isogenic wheat lines deficient in the individual HMW‐GS will allow the determination of the contributions of the different alleles of each locus to dough quality.
