Regulated expression of glutamyl‐tRNA synthetase is directed by a mobile genetic element in the cyanobacterium Tolypothrix sp. PCC 7601

Abstract: SummaryThe genome of Tolypothrix sp. PCC 7601 carries two copies of a novel insertion sequence, IS Tosp 1. One of the two copies is located upstream of the gene encoding glutamyl-tRNA synthetase, an enzyme playing a key role in protein and pigment synthesis. The tnpA gene of the IS element and gltX were co-transcribed and their expression was transiently upregulated upon retrieval of the ammonium source irrespective of whether nitrate or no nitrogen source were available. The second copy is also transcribed an… Show more

“…Although IS elements are typically associated with genetic rearrangements, other functions have been reported, including service as promoter elements for nearby genes (24). There are, however, no reports of the gene regulation in Francisella observed in the present study occurring on a global scale (23,24,30). Moreover, this is the first report of polyamines regulating transcription from IS elements.…”

“…Moreover, as one of aminoacyl-tRNA synthetases, glutamyl-tRNA synthetase can participate in a wide variety of biological processes including RNA processing and trafficking, apoptosis, transcription of rRNA, angiogenesis, and inflammation (Lee et al 2004;Park et al 2005). Recently, structure and regulated expression of glutamyltRNA synthetase were further presented (Levican et al 2005;Luque et al 2006;Schulze et al 2006). Although a large number of studies on glutamyl-tRNA synthetase were reported, almost all these studies focused on microorganisms and animals and knowledge of plant glutamyl-tRNA synthetase remains quite limited.…”

“…Western blots were performed as described previously (29). Antibodies against an internal peptide (EYDRKKDESPSVEAC) of Anabaena ValRS were obtained from rabbits and affinity purified (Genscript, Piscataway, NJ).…”
