Regulation of bone matrix protein expression and induction of differentiation of human osteoblasts and human bone marrow stromal cells by bone morphogenetic protein-2

Abstract: We have examined the effects of BMP-2 on the expression of bone matrix proteins in both human bone marrow stromal cells (HBMSC) and human osteoblasts (HOB) and their proliferation and mineralization. Both HBMSC and HOB express BMP-2/-4 type I and type II receptors. Treatment of these two cell types with BMP-2 for 4 weeks in the presence of beta-glycerophosphate and ascorbic acid results in mineralization of their matrix. BMP-2 increases the mRNA level and activities of alkaline phosphatase and elevates the mRN… Show more

“…For valvular calcification studies, digital photomicrographs were captured with a Spot Enhanced camera mounted on a Zeiss Axiovert S100 microscope. Mineral deposition was visualized with Alizarin red (calcium) (29) or von Kossa (phosphate) (15) stains, and the mineralized leaflet area was quantified using Kodak one-dimensional automated image analysis software version 3.5.3. The data are expressed as the percentage of mineralized area of total valve leaflet area (pixel counts) from digital photomicrograph images, averaging results over five adjacent 5-micron sections for each animal (n ϭ 5-6 animals/treatment arm).…”

“…Human aortic smooth muscle cell cultures isolated from atherosclerotic aortas demonstrated a BMP-2-and -4 -activated osteoblastic gene transcription program. 23 Phosphorus stimulated mineralization of the extracellular matrix (ECM) of these cultures analogous to bone formation in vitro. Mineralization was blocked by a BMP antagonist, noggin, in the presence of high phosphorus.…”

“…TGFβ 1 (25 kDa) influences the proliferation and differentiation of the stem cells (28), while BMP 2 (26 kDa) stimulates the production of specific bone matrix proteins (29,30). The proteins were embedded within a film of alternating layers of the polypeptides poly-L-lysine (PLL) and poly-L-glutamic acid (PGA) (31,32).…”
