BackgroundTomato fruit ripening is controlled by ethylene and is characterized by a shift in color from green to red, a strong accumulation of lycopene, and a decrease in β-xanthophylls and chlorophylls. The role of other hormones, such as auxin, has been less studied. Auxin is retarding the fruit ripening. In tomato, there is no study of the carotenoid content and related transcript after treatment with auxin.ResultsWe followed the effects of application of various hormone-like substances to “Mature-Green” fruits. Application of an ethylene precursor (ACC) or of an auxin antagonist (PCIB) to tomato fruits accelerated the color shift, the accumulation of lycopene, α-, β-, and δ-carotenes and the disappearance of β-xanthophylls and chlorophyll b. By contrast, application of auxin (IAA) delayed the color shift, the lycopene accumulation and the decrease of chlorophyll a. Combined application of IAA + ACC led to an intermediate phenotype. The levels of transcripts coding for carotenoid biosynthesis enzymes, for the ripening regulator Rin, for chlorophyllase, and the levels of ethylene and abscisic acid (ABA) were monitored in the treated fruits. Correlation network analyses suggest that ABA, may also be a key regulator of several responses to auxin and ethylene treatments.ConclusionsThe results suggest that IAA retards tomato ripening by affecting a set of (i) key regulators, such as Rin, ethylene and ABA, and (ii) key effectors, such as genes for lycopene and β-xanthophyll biosynthesis and for chlorophyll degradation.Electronic supplementary materialThe online version of this article (doi:10.1186/s12870-015-0495-4) contains supplementary material, which is available to authorized users.

The rice height has been proposed to involve gibberellin biosynthesis. In order to regulate rice height, we suppressed expression of OsGA20ox2 by the double-stranded RNA interference (RNAi) method. For this purpose, we constructed two different RNAi vectors, pCH1CK and pCH12CK, then performed Agrobacterium-mediated transformation of rice varieties QX1 and Zhongzuo0201. The RNAi transgenic lines derived from pCH1CK and pCH12CK demonstrated a clear reduction in height compared with the control plants (P<0.01). The height of dwarf lines from pCH1CK and pCH12CK were about 75-84% and 54-74% of that of the control plants, respectively. Transcripts for semi-dwarf lines from pCH1CK and pCH12CK were also reduced in comparison with the control plants. Analyses of seeds per panicle, 1,000-grain weight, and seed yield per plant suggested that the RNAi semi-dwarf lines showed stable grain yield compared with the wild-type plants. We further showed that the RNAi semi-dwarf lines could be restored to normal plant height by applying exogenous GA 3 . The results also indicate that different fragment of the same RNAi target can be practically applied to produce different extent semi-dwarf of rice plants.

Carotenoid Accumulation and Carotenogenic Genes Expression During Two Types of Persimmon Fruit (Diospyros kaki L.) Development

Persimmon (Diospyros kaki L.), belonging to the Ebenaceae family, has been used not only as a fresh fruit, but also for many medicinal uses. Carotenoids are the main pigment in persimmon fruit, which contribute significantly to fruit color and nutritional quality due to their composition and content. In this study, fruit quality indices, carotenoid contents and expression of carotenogenic genes were analyzed in two types of persimmon fruit. The results demonstrated that there was a positive correlation between fruit color and the contents of main composition and total carotenoids. Carotenoid accumulation in persimmon fruit resulted from the interaction of carotenogenic genes, but the molecular mechanisms responsible for accumulation of carotenoids in two types of persimmon fruit had a few differences. As a complete unit, the relatively low expression level of phytoene synthase gene (DkPSY) in "Niuxinshi" resulted in low carotenoid contents or even under the detection limit at the early fruit developmental stages; but low carotenoid contents in "Nishimurawase" were due to the relatively low expression level of carotenogenic genes other than DkPSY. At the late fruit developmental stages, increased expression levels of DkPSY, phytoene desaturase gene and beta-carotene hydroxylase gene (DkBCH) induced elevated carotenoid contents; because all carotenogenic genes strongly expressed in "Nishimurawase", a large amount of carotenoids were accumulated. In addition, β-cryptoxanthin was the main composition whose content increased with the fruit maturity changes, which was mainly because of DkBCH which might lead more conversion of β-carotene to β-cryptoxanthin.
