Regulation of differentiation of sheep subcutaneous and abdominal preadipocytes in culture

Abstract: Factors regulating the differentiation of sheep subcutaneous and abdominal (omental or perirenal) preadipocytes from foetal lambs, suckling lambs and fattening sheep have been investigated using a serum-free cell culture system. Differentiation was assessed by changes in the activity of the enzyme glycerol 3-phosphate dehydrogenase. Insulin or IGF-I was essential for differentiation. Dexamethasone, a lipid supplement (Excyte) and the thiazolidinedione, rosiglitazone (BRL 49653) (a peroxisome proliferator-activ… Show more

“…This is consistent with there being depot-specific responses of preadipocytes to exogenous factors that regulate preadipocyte differentiation (Soret et al 1999). In our study, PR adipose tissue was susceptible to the programming effects of maternal nutrient intake during the first and second trimesters of gestation, whereas OM adipose tissue was only susceptible during the second trimester.…”

“…This is supported by the central role of PPARg in the control of the terminal differentiation of pre-adipocytes isolated from the stromalvascular fraction in bovine WAT Torii et al, 1998) and ovine BAT (Soret et al, 1999). However, CEBP-a expression was either not detected or did not parallel the adipogenic process (Ohsaki et al, 2007) in cattle.…”

“…Their interest was demonstrated by the fact that their adipose conversion process exhibits some differences when compared to that of cell lines. These studies were essentially performed with rat preadipose cells but preadipocytes from murine, rabbit, ovine, porcine, bovine and human species were also successfully cultured [27,67,92,107,113,152,174,182,216]. These cells can be isolated from different fat depots, as well as from animals of different ages and/or different physiological state [92,104,139].…”
