Regulation of hypothalamic gene expression by glucocorticoid: implications for energy homeostasis

Abstract: Using the serial analysis of gene expression (SAGE) method, we studied the effects of adrenalectomy (ADX) and GC on the transcriptomes of mouse hypothalamus. Approximately 180,000 SAGE tags, which correspond to 50,000 tag species, were isolated from each group of intact or adrenalectomized mice as well as 1, 3, and 24 h after GC injection. ADX upregulated diazepam binding inhibitor gene expression while downregulating vomeronasal 1 receptor D4, genes involved in mitochondrial phosphorylation (cytochrome-c oxid… Show more

“…GSE333-66 10 contains expression data from MCF-7 tumor xenografts treated with tamoxifen or nothing for 28 d, with each group having 2 biological replicates. GSE26459 11 contains expression data from subcloned MCF-7 cell lines that were either naturally resistant or highly sensitive to tamoxifen, with each group having 3 biological replicates.…”

“…Using serial analysis of gene expression (SAGE) strategy which indicates the relative level of expression of each transcript matched to a tag, this study represents, to our knowledge, the first characterization of the tissue-specific genes and housekeeping genes in an extensive array of tissues and organs. Using SAGE method, previous studies have characterized the transcriptome of numerous tissues such as uterus [7], adipose tissue [8,9], skeletal muscle [10], hypothalamus, cerebral cortex and pituitary gland [11,12]. …”

“…A similar phenomenon of presence of weight gain with rising total leptin level was described in humans with glucocorticoid excess—who gain weight and have good appetite26 despite rising total leptin levels 27. Glucocorticoids affect energy storage28 via decreased thermogenesis,29 increased lipogenesis (with increased insulin levels)30,31 and altered central neural regulation of energy homeostasis 32…”
