Regulation of Inflammatory Reaction in Health and Disease

Abstract: Inflammation is a key mechanism for the clearance of infective agents and other inflammatory triggers and is pivotal for the repairing processes of the affected tissues. Inflammation is a multistep process driven by a great number of mediators which regulate specific aspects of the inflammatory response, in agreement with a well-defined chronobiological program. A great number of inflammation-related diseases show a deeply altered immune chronobiology (e.g., COVID-19-related cytokines storm). This aspect highl… Show more

“…Some proteins, such as β-catenin and COX-2, are closely associated with wound healing ( Li et al, 2016 ; Chen et al, 2017 ). Inflammation is an integrative process driven by a number of mediators which regulate specific aspects of the inflammatory response ( Fioranelli et al, 2021 ). Under normal conditions, inflammatory reactions will promote the proliferation of adult fibroblasts ( Carre et al, 2010 ), up-regulate the production of β-catenin ( Schunk et al, 2021 ), COX-2, and the secretion of prostaglandins ( Enzerink et al, 2010 ), activate vascular endothelial growth factors (VEGFs), and promote wound healing by facilitating wound angiogenesis and accelerating blood supply ( de Oliveira et al, 2014 ).…”

“…The initiation of a specific immune response depends on the recognition of the foreign nature of an antigen as well as the conditions under which the given antigen is presented to immunocompetent cells: if the conditions are interpreted by the organism as pathological, they will trigger an immune response regardless of whether the antigen is the organism's own or foreign [87]. In the case of auto-aggression, the signal inducing the immune response may originate from an inflammation accompanying the release of antigens due to damage [88]. The immunological markers of type 1 diabetes include antibodies active against pancreatic β cells: Islet-cell antibodies (ICA), insulin autoantibodies (IAA), anti-glutamine acid decarboxylase (anti-GAD), anti-zinc transporter protein 8 (anti-ZnT8), and anti-tyrosine phosphatase antibodies (anti-IA2) [89].…”

“…In some situations, humans lose the capacity to control this reaction and its long-term existence results in a chronic systemic inflammatory state [ 87 ]. Uncontrolled and prolonged inflammatory processes are implicated in the etiology of many chronic conditions, such as cancer, asthma, diabetes mellitus, chronic kidney, cardiovascular, osteo-degenerative, and non-alcoholic fatty liver diseases as well as autoimmune and neurodegenerative disorders [ 88 , 89 , 90 ]. To abrogate the burden of inflammatory diseases in man, a number of interventions are already in place, such as the use of anti-inflammatory drugs and natural compounds [ 91 , 92 ].…”
