Regulation of the Actin Cytoskeleton by an Interaction of IQGAP Related Protein GAPA with Filamin and Cortexillin I

Abstract: Filamin and Cortexillin are F-actin crosslinking proteins in Dictyostelium discoideum allowing actin filaments to form three-dimensional networks. GAPA, an IQGAP related protein, is required for cytokinesis and localizes to the cleavage furrow during cytokinesis. Here we describe a novel interaction with Filamin which is required for cytokinesis and regulation of the F-actin content. The interaction occurs through the actin binding domain of Filamin and the GRD domain of GAPA. A similar interaction takes place… Show more

“…This stress-dependent myosin II accumulation results from cooperative interactions between the actin-bound myosin heads and the actin-bundling protein cortexillin I [4]. Scaffolding proteins IQGAP1 and IQGAP2 bind to cortexillin I [1214] and regulate myosin II accumulation [6]. IQGAP1 inhibits myosin II recruitment, while IQGAP2 relieves this inhibition.…”

“…Because IQGAP1 and IQGAP2 interact with distinct domains of cortexillin I [1214], we also studied cortexillin I dynamics in the iqg1/2 double mutant. Here, cortexillin I still showed faster recovery at the furrow as compared to WT, similar to iqg2 (Fig.…”

“…It promotes actin ring formation by binding to actin filaments via its CHD (Calponin Homology Domain) [24, 25, 27, 76, 112], and it is required for the localization or maintenance of Myo1 at the division site during cytokinesis [25]. Both roles appear conserved in fission yeast [86, 87, 89, 113] and Dictyostelium [88, 109, 114]. …”

“…Several proteins have been identified as cortexillin binding partners. Cortexillin I interacts with RacI, IQGAPs, and GAPA forming a complex, which plays a key role in cytokinesis and cell migration Mondal et al, 2010). Furthermore, an interaction with Rap-GAP1 has been demonstrated.…”

“…The last positively charged nonapeptide motif has a phosphatidylinositol (4,5)bisphosphate (PIP2)-binding site . Cortexillin I forms a complex with RacI and the IQGAPs DGAP1 and GAPA (Faix et al, 2001;Mondal et al, 2010). The IQGAP/cortexillin complexes are involved in regulating cortical mechanics.…”
