Release of Peroxidases by Cultured Potato Cells

Abstract: Electrophoretic analysis of peroxidase isoenzymes and other proteins present in the culture medium of potato (Solanum tuberosum L. cv. Bintje) cells in suspension culture was carried out by isoelectric focusing and starch gel electrophoresis. The results showed that during the exponential growth phase, the number of peroxidase isoenzymes as well as their activities increased. In addition, several other proteins have increased in the culture medium. The high specific activity of medium peroxidase as compared to … Show more

“…Cell suspensions from two or three flasks per time were pooled and then cells were collected by suction on a Buchner funnel. The fresh weight of the cells was determined and one gram of sample was used for extraction as described previously (BREDEMEIJER et al 1985). The culture filtrates were only centrifuged (18,000 g for 30 min) and used without further treatments for peroxidase assay and electrophoresis.…”

“…In cultures of lupin [29] and potato cells [8], the occurrence of dehydrogenases was similarly restricted to the cell saps. Moreover, the time courses of the total activities as recorded for the in vitro cultured apple cells are different for cell saps and media, e.g.…”

“…Despite these early findings and in contrast to microbial and animal cell cultures, more recent studies on the distribution of enzymic activities in plant cell cultures are limited [10,29,30]. Several investigators have differentiated the extracellular activities with electrophoretic methods [1,3,5,8,19], but, with two exceptions [1,19], these studies were restricted to peroxidase isozymes. It is, however, doubtful whether the peroxidase system is the most suitable one, especially in physiological studies, because of its multifunctional character and its susceptibility to many environmental changes [24,27].…”

“…1A and 1B). Extracellular peroxidase has been reported in suspension cultures of carrot [9], peanut [10], potatoes [11], spinach [12], cotton [13] and horseradish [1]. In the case of petunia, the peroxidase secreted into the extracellular medium can reach 15% of the total secreted protein [10].…”
