Micro/nano engineering is an emerging field which enables engineering and scientific discoveries in the microworld. As an effective and powerful tool for automation and manipulation at small scales, precision motion measurement by computer micro-vision is now broadly accepted and widely used in micro/nano engineering. Unlike other measurement methods, the vision-based techniques can intuitively visualize the measuring process with high interactivity, expansibility, and flexibility. This review paper aims to comprehensively present a survey of micro-vision-based motion measurement from the collective experience. Working principles of micro-vision systems are firstly introduced and described, where the hardware configuration, model calibration, and motion measurement algorithms are systematically summarized. The characteristics and performances of different micro-vision-based methods are then analyzed and discussed in terms of measurement resolution, range, degree of freedom, efficiency, and error sources. Recent advances of applications empowered by the developed computer micro-visionbased methods are also presented. The review can be helpful to researchers who engage in the development of micro-vision-based techniques, as well as provide the recent state and tendency for the research community of vision-based measurement, manipulation, and automation at micro/nano scales.

This paper presents the design and development of a new microinjection system with force sensing for automated multi-cell injection in genome editing of zebrafish using the CRISPR/Cas9 system. In comparison with the traditional approach, it achieves a high injection speed and high precision of operation, which leads to consistent quality and high survival rate of the injected cells. One unique feature is that force sensing is employed to determine the exact moment when the cell membrane is pierced, which contributes to the protection of the cell from damage. Moreover, a new cell holding device is designed to immobilize the cells, which speeds up the multi-cell microinjection process by eliminating the use of conventional vacuum holding pipette. The system implements the object detection by machine vision algorithm along with position/force regulation. The machine vision is realized by combining the pyramid template matching algorithm with Kalman filter, which reduces the computation time and facilitates real-time control. The experimental studies are conducted to verify the performance of the developed system. Results demonstrate the reliability of the robotic microinjection system, which offers a more consistent injection quality and higher viability of cells over the skilled human operator.

Design and implementation of CCD image-based DNA chip scanner with automatic focus calibration

To address the slow scanning speed of the laser confocal method for scanning DNA chips, we designed and built a CCD image-based DNA chip scanner. A high-powered LED was selected as excitation light source. To avoid stray light in the optical path, we designed a light entrance, an extinction dark room, a diaphragm, and an extinctor that eliminated 99.95% of the stray light. A focal automatic calibration scheme is proposed; the only addition to the hardware was the focusing motor and the connection device between the motor and the lens. Furthermore, variance was used as the image sharpness evaluation function, with the combination of rough and fine calibration to increase real-time performance. An experiment showed that the accuracy of automatic focal calibration is higher than that of the manual method with repeatability error of 2.1%, and the time required is between 18.72 and 55.78 s. The sensitivity can reach 1.12 flour/μm 2 with a resolution of 10.04 μm. Using the proposed scanner and a commercial Boao scanner on a fluorescence dot on the same genetic defect chip found a consistency coefficient of 0.95. The scanner has the advantage of simple hardware structure, quick detection, and favourite performance.
