In the growing seasons from 2003 to 2008, 547 isolates of Phytophthora infestans from five regions in the Czech Republic were collected and examined for their sensitivity to the active ingredients (metalaxyl, dimethomorph and propamocarb-HCl) of frequently used fungicides. The response of the isolates to each of these substances was examined using the in vitro amended-agar method; in 352 of these isolates, the sensitivity to metalaxyl was also assessed by the floating leaf-disc assay. The majority of the isolates were sensitive (89.8%) to metalaxyl. All the isolates that were tested were sensitive to dimethomorph and propamocarb-HCl; these particular substances completely suppressed mycelial growth at 1 µg a.i. per ml.

There is limited understanding of the genetic variability in Phytophthora infestans in the major potato cultivation region of north-western Russia, where potato is grown primarily by small households with limited chemical treatment of late blight. In this study, the mating type, sensitivity to metalaxyl, and genotype and population genetic diversity (based on 12 simple sequence repeat (SSR) markers) of 238 isolates of P. infestans from the Pskov region during the years 2010−2013 were characterized. The aim was to examine the population structure, phenotypic and genotypic diversity, and the prevalent reproductive mode of P. infestans, as well as the influence of the location, time, and agricultural management practices on the pathogen population. The frequency of the A2 mating was stable over the four seasons and ranged from 33 to 48% of the sampled population. Both mating types occurred simultaneously in 90% of studied fields, suggesting the presence of sexual reproduction and oospore production in P. infestans in the Pskov region. Metalaxyl-sensitive isolates prevailed in all four years (72%), however, significantly fewer sensitive isolates were found in samples from large-scale conventional fields. A total of 50 alleles were detected in the 141 P. infestans isolates analyzed for genetic diversity. Amongst the 83 SSR multilocus genotypes (MLGs) detected, 65% were unique and the number of MLGs varied between locations from 3 to 20. These results, together with the high genotypic diversity observed in all the locations and the lack of significance of linkage disequilibrium, suggest that sexual recombination is likely responsible for the unique MLGs and the high genetic diversity found in the Pskov region population, resembling those of north-eastern European populations.

A B S T R A C T Phytophthora infestans, the causal agent of potato and tomato late blight disease continues to be a serious threat in almost all potato and tomato growing areas around the world, with dramatic and disastrous economic consequences. In Mauritius, the disease is reported every year and many of the commercial potato varieties (Spunta, Delaware and Mondial) previously found to be moderately resistant or susceptible to the disease, have now become highly susceptible. This study reports the first phenotypic and genotypic characterization of local strains of Phytophthora infestans from the island of Mauritius. Seven isolates of Phytophthora infestans were collected from distinct geographical areas during the blight seasons of 2012 and 2013 and tested for aggressiveness on four potato and three tomato cultivars, metalaxyl sensitivity and subjected to isozyme analysis at the Gpi and Pep loci. Results show that all the isolates exhibited a clear host adaptation to potato, being only very weakly pathogenic on tomato. Most of the isolates (five out of seven) had high or intermediate metalaxyl resistance. Isozyme analysis revealed that at the Gpi locus, two isolates were 86/86, one isolate was 86/100, one isolate was 100/100 and one isolate was 122/122. At the Pep locus, two isolates were 92/92, two isolates were 92/100 and three isolates were 100/100. The combined results of the tests indicate that the seven isolates collected over the island are genetically distinct from each other and do not represent the same strain.
