Response of Thyroid Follicular Cells to Gamma Irradiation Compared to Proton Irradiation. I. Initial Characterization of DNA Damage, Micronucleus Formation, Apoptosis, Cell Survival, and Cell Cycle Phase Redistribution

Abstract: The RBE of protons has been assumed to be equivalent to that of photons. The objective of this study was to determine whether radiation-induced DNA and chromosome damage, apoptosis, cell killing and cell cycling in organized epithelial cells was influenced by radiation quality. Thyroid-stimulating hormone-dependent Fischer rat thyroid cells, established as follicles, were exposed to gamma rays or proton beams delivered acutely over a range of physical doses. Gamma-irradiated cells were able to repair DNA damag… Show more

“…Typically, an increase in LET has been associated with altered biological effects, including unique gene expression patterns (Green et al, 2001;Woloshak, 1997) and impairment of conditioned taste aversion (Rabin et al, 1989;Rabin et al, 1991). In most models, an increase in LET results in a decrease in behavioral performance.…”

“…Femoral bone marrow was isolated from CBA/H and C57BL/6 strains. Bulk cultures containing cells surviving varying doses of 250 keV X-irradiation were analysed for chromosome damage (as in Kadhim et al, 1999) and apoptosis (as in Green et al, 2001). The first three data sets along the x-axis are CBA/H, the second three are C57BL/6.…”

“…Bone marrow was isolated after whole-body irradiation with 250 keV 60 Co g-irradiation, and analysed for % apoptotic cells at days 1 and 365 using the Annexin V assay (Green et al, 2001). Per cent apoptotic cells are normalized for control levels.…”

“…The cells used in these experiments were derived from our pass 3 through pass 25 FRTL-5 stocks. Our care and feeding of these cells has been previously described (see Green et al 2001). For clarification, Ham's modified F-12 is the medium specified by ATCC, we however, maintain our FRTL-5 cells in DMEM:F12 (50:50 v/v).…”

“…Follicle counts were performed as previously described in Green et al (2001). Briefly, plates/wells from which the supernatants were harvested were fixed with methanol:acetic acid (3:1 v/v) for 15 min at room temperature followed by crystal violet staining.…”
