Foot-and-mouth disease (FMD) is a highly contagious disease affecting ruminants and pigs. In countries in which control of FMD relies predominantly on vaccination, young stock ingest specific anti-FMD virus antibodies in the colostrum. This maternally-derived antibody (MDA) provides immediate protection against infection with FMD virus, but also interferes with the development of active immunity following vaccination. However, susceptibility to infection precedes the ability to respond to vaccination in the presence of MDA. Currently available vaccines cannot overcome this inhibitory effect of MDA, and protection of young stock can only be provided by their isolation from FMD virus.

Data from our laboratory and others show that calves are immunologically identical to adult animals at this age (Sadir et al 1988;Spath et al 1995;Bucafusco et al 2014); thus, we do not expect an impact on antibody titres due to this bias. We cannot rule out, however, the role of IgMs, Nabs or high-affinity IgGs as a result of multiple vaccinations in the results obtained by either test.…”

Buffaloes are compulsory vaccinated against foot-and-mouth disease virus (FMDV) in many countries as part of the official control programmes. Serological testing aimed to indirectly assess herd immunity is currently performed using the same Enzyme-linked immunosorbent assays (ELISAs) applied for bovine sera, assuming an agreement between the ELISA's diagnostic results and those obtained using the virus neutralization test (VNT). Here we evaluated the accuracy of different ELISA tests to assess vaccine-induced antibodies against FMDV in buffalo's sera classified according to their VNT titres. Currently used liquid-phase blocking ELISA yielded very low specificity, producing high titres for many samples with low VNT titres. To increase specificity, we developed an indirect ELISA using purified 140S viral particles and an avidity single-dilution ELISA, which includes a urea washing step after the incubation of the diluted serum sample, to detach weak binders. Combining these two highthroughput single-dilution tests, an excellent concordance with VNT was achieved. This is the first study analysing the diagnostic agreement of traditional and novel serological tests with VNT for the indirect assessment of antibodies against FMDV capsid proteins in buffalo serum samples. The influence of age and maternal antibodies on the development and duration of postvaccinal antibody response against Glässer's disease were investigated. Pigs born to immune (MDA-positive) and non-immune (MDA-negative) sows were vaccinated with inactivated vaccine. Vaccination was done according to three different protocols: at 1 and 4, at 2 and 5 or at 4 and 7 weeks of age. There were also two control groups for MDA-negative and MDA-positive pigs. The level of Haemophilus parasuis (Hps) specific antibodies were determined using commercial ELISA test. No serological responses were seen in any of the groups after the first vaccination. Maternally derived antibodies (MDA) against Hps were above the positive level until approximately 3 weeks of life in MDA-positive pigs. In those pigs the strongest postvaccinal humoral response was observed in piglets vaccinated at 4 and 7 weeks of age. In the remaining MDA-positive piglets only slight seroconversion was noted but levels of antibodies never exceeded values considered as positive. All MDA-negative pigs produced Hps-specific antibodies after the second vaccination. The results of the present study indicated that MDA may alter the development and duration of active postvaccinal antibody response. Age of pigs at the moment of vaccination was not associated with the significant differences in the magnitude of antibody response, however influenced the kinetics of decline of Hps-specific antibodies.
