Restricted junctional usage of T cell receptor Vβ2 and Vβ13 genes, which are overrepresented on infiltrating T cells in the lips of patients with sjögren's syndrome

Abstract: Objective. To analyze the clonality of T cell receptor (TCR) Vβ2—and Vβ13—positive T cells, which are predominantly expressed in the lips of patients with Sjögren's syndrome (SS).Methods. The junctional sequences of complementary DNA clones encoding TCR Vβ2 and Vβ13 genes were determined by the polymerase chain reaction. Forty‐one Vβ2 and 45 Vβ13 clones established from the lips of 3 SS patients were sequenced.Results. The Vβ2/Jβ2.3 pair was enriched in 2 of the 3 patients (44% and 46% of the clones, respectiv… Show more

“…Moreover, sequence analysis of the complementary-determining region 3 has unveiled certain conserved amino acid motifs. These observations support the notion that infiltrating T cells recognize relatively few epitopes on the autoAg [1][2][3][4]. AutoAgs recognized by T cells that have infiltrated the SGs have been analyzed.…”

“…These results indicate that the M3R immune reaction plays a key pathogenic role in AIS, suggesting that M3R molecule acts as an autoantigen in the pathogenesis of SS. [1,2] and Va genes on T cells present in the SGs, LGs, kidneys and peripheral blood (PB) of patients with SS, suggesting a preferential selection of TCR genes. Moreover, sequence analysis of the complementary-determining region 3 has unveiled certain conserved amino acid motifs.…”

“…26,27 Patterns of restricted TCR gene expression have also been described for a variety of human autoimmune diseases, [28][29][30] including inflammatory myopathies. [14][15][16][17]19,23,[30][31][32][33][34][35][36][37][38] The significantly higher degree of clonal expansion of T cells seen in our sIBM muscle samples, compared with their corresponding PBL, suggests that the T cells are specifically recruited to the muscle or expand in situ, rather than by passive recirculation between blood and muscle.…”

“…However, since most T cell responses to complex antigens involve multiple peptides, it is hard to explain the data of Yumoto et al [21] in this context. Interestingly, a study of TCR V]3 usage in Sj6gren's disease, a condition known to be a precursor of MALT-type lymphomas in the salivary gland showed dominance of V~32 and VI313 gene expression [14,20]. In a study of TCR Vc~ and V~3 gene usage by T cells infiltrating the thyroids of patients with autoimmune thyroid disease, including 4 patients with Hashimoto's thyroiditis from which thyroid lymphomas of MALT-type arise, unrestricted TCR V~ but a restricted Vc~ gene usage was observed [3].…”
