Results of a 5 Years Prospective Study of Estriol Succinate Treatment in Patients with Climacteric Complaints

Abstract: In a prospective study 911 patients were treated over a period of 5 years (M = 2.2) or a total of 2007 treatment years with estriol succinate oral (Synapause, 2-12 mg per day). The treatment was very effective in the removal of all typical climacteric complaints and of the atrophic genital changes caused by estrogen deficiency. Subjective side effects were seldom seen and without practical importance for the treatment. Objective, grave side effects were only few: one superficial phlebo-thrombosis, 2 cases of t… Show more

“…symptoms over 40 years. [11][12][13] The standard daily dose for menopause symptoms (2 mg) is generally lower than the dose we used, although some studies have used up to 16 mg. In this study, a dose of 8 mg induced an estriol concentration equivalent to that present early in the second trimester of pregnancy, consistent with previous fi ndings.…”

“…[7][8][9] Estriol has been used for several decades throughout Europe and Asia for treating menopausal symptoms. [11][12][13] A Women's Health Initiative study 14 done in 2002 to assess whether premarin (a complex mix of conjugated oestrogens) protects against coronary heart disease in menopausal women aged 50-79 years was stopped prematurely because of an increased risk for cardiovascular disease and breast cancer, whereas the number of colorectal cancers and hip fractures decreased. Premarin includes estradiol, an oestrogen that is present at low concentrations in women with normal menstrual cycles and in oral contraceptives.…”

“…than benefit in preventative medicine strategies in healthy menopausal women, in whom only minimum to no toxicity is tolerable, the risk/benefit ratio in patients with a chronic autoimmune disease is quite different, with modest toxicity tolerable. The use of pregnancy doses of estriol, the safest of the three estrogens (63)(64)(65), may provide an opportunity to optimize anti-inflammatory efficacy and minimize toxicity.…”
