The knowledge of the genetic diversity of barley (Hordeum vulgare) genotypes based on protein polymorphism is very important for breeding programs. The purpose of the current study was to determine the genetic diversity and relationships among ten barley genotypes by using Sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) for protein profiles. A total number of 30 bands with molecular weights ranging from 12 to 148 KD were detected. Out of these, five bands were observed monomorphic. Rest of the bands had shown polymorphism to the extent of 83.3% among the test genotypes. The genetic similarity of the ten genotypes tested varied from 0.26 to 1.00 with an average of 0.51. Cluster analysis divided the ten genotypes into two major clusters comprising four subclusters, which was consistent with the systematic classification of barley done in previous studies. The results of this study indicated that the genotypes of barley could effectively be differentiated based on polymorphism, detected between protein patterns. SDS-PAGE presented a higher differentiation power and better repeatability; thus, could be used as a rapid and reliable method for genetic diversity analysis and laid a solid foundation for future barley breeding.

Total seed storage proteins were studied in 50 accessions of (11 ssp. var, 13 ssp. var , 11 ssp. var and 15 ssp. var. accessions) in SDS PAGE. These accessions were also analysed for albumin and globulin seed protein fractions. Among the six seed protein markers presently used, it was found that globulin fraction showed maximum diversity (77.2%) in accessions followed by albumin (52.3%), denatured total soluble protein fraction in embryo (33.3%) and cotyledon (28.5%). The cluster analysis based on combined data of cotyledons, embryos, albumins and globulins seed protein fractions demarcated the accessions of two subspecies and into two separate clusters supported by 51% bootstrap value, with few exceptions, suggesting the genotypes to be moderately diverse. Native and denatured total soluble seed storage proteins were also electrophoretically analysed in 27 wild species belonging to six sections of the genus. Cluster analysis using different methods were performed for different seed proteins data alone and also in combination. Section Caulorrhizae (C genome) and Triseminatae (T genome) formed one, distantly related group to and other section Arachis species in the dendrogram based on denatured seed storage proteins data. The present analysis has maintained that the section Arachis species belong to primary and secondary genepools and, sections Procumbenetes and Erectoides belong to tertiary gene pools.
