Reversal of muscle atrophy by Zhimu and Huangbai herb pair via activation of IGF-1/Akt and autophagy signal in cancer cachexia

Abstract: The results revealed that ZBHP treatment could inhibit the muscle atrophy induced by cancer cachexia and prolong the survival time, and ZBHP may be of value as a pharmacological alternative in treatment of cancer cachexia.

“…A decrease in body weight and the skeletal muscle weight are the main symptoms of patients with cancer cachexia and are also the symptoms observed in the experimental C26 mouse model of cancer cachexia [30]. The reduction in the tumor-free body weight, epididymal fat and gastrocnemius muscles indicated that the model of cancer cachexia mice was successfully established by injecting C26 mouse colon cancer cells into BALB/c mice.…”

“…Recently, several herbal medicines, including Hochuekkito (TJ-41) and Coptidis rhizoma , have been demonstrated to attenuate cancer cachexia by reducing IL-6 production, which is a critical mediator of muscle wasting [ 39 , 40 ]. In addition, ZBHP, comprising the herbs Rhizoma Anemarrheana and Cortex Phellodendri , increased body weight and suppressed tumor-induced muscle atrophy by inhibiting pro-inflammatory cytokines and muscle catabolism in a C26 model [ 19 ]. In our previous studies, Citrus unshiu peel extract and the Oriental traditional herbal formula Sosiho-tang (Xiaochaihu-tang in Chinese, Sho-saiko-to in Japanese) efficiently improved cachexia-related symptoms in CT-26 tumor-bearing mice by reducing systemic inflammation and muscle degradation [ 32 , 41 ].…”

“…In addition, antibodies or synthetic peptides targeting cachectic mediators have been effective in reversing cachexia conditions [ 15 , 16 ]; however, these agents have a high cost and lack of clinical data for their effectiveness as well as safety. Recently, herbal medicines have proven to be beneficial for managing cancer-induced cachexia symptoms, including anorexia, weight loss, fatigue, and muscle wasting, in tumor-bearing mice because of their multi-modal pharmacological actions and low toxicity [ 17 19 ].…”
