Reverse Genetics Systems for Emerging and Re-Emerging Swine Coronaviruses and Applications

Hui Jiang,
Ting Wang,
Lingbao Kong
et al.

Abstract: Emerging and re-emerging swine coronaviruses (CoVs), including porcine epidemic diarrhea virus (PEDV), porcine deltacoronavirus (PDCoV), and swine acute diarrhea syndrome-CoV (SADS-CoV), cause severe diarrhea in neonatal piglets, and CoV infection is associated with significant economic losses for the swine industry worldwide. Reverse genetics systems realize the manipulation of RNA virus genome and facilitate the development of new vaccines. Thus far, five reverse genetics approaches have been successfully ap… Show more

“…And these RGSs have been used to study the functions of several PEDV structural and nonstructural proteins and in generating PEDV LAV candidates [50,80,104,114,140,155]. Recently, Jiang et al reviewed the principles, advantages, and drawbacks of the established RGSs for the emerging and re-emerging swine coronaviruses, including PEDV [156]. Thus, we will only provide a brief overview of PEDVs' RGSs.…”

“…Except for retroviruses, replicons derived from positive-sense RNA viruses do not integrate exogenous genetic information into the host cell genome. Many systems have been described for heterologous gene expression based on infectious cDNA clones of filoviruses [ 7 ], influenza viruses [ 8 ] (reviewed in [ 9 ]), reoviruses [ 10 , 11 , 12 ], rotaviruses [ 13 , 14 ] (reviewed in [ 15 ]), bornaviruses [ 16 ], bunyaviruses [ 17 ], picornaviruses [ 18 ], flaviviruses [ 19 ], alphaviruses [ 20 ], and coronaviruses [ 21 , 22 , 23 , 24 ], among others.…”
