Rheumatoid Arthritis Fibroblast-like Synoviocyte Suppression Mediated by PTEN Involves Survivin Gene Silencing

Abstract: Survivin is a proto-oncogene biomarker known for its anti-apoptotic and cell cycle regulating properties induced by the activation of the phosphoinositide 3-kinase (PI3K)/Akt pathway. In the context of non-cancer pathology, such as rheumatoid arthritis (RA), survivin has emerged as a feature associated with severe joint damage and poor treatment response. Phosphatase and tensin homolog (PTEN) is a phosphatase antagonizing all classes of PI3K. The interplay between survivin oncogenic mechanisms and proliferatio… Show more

“…| 21579 study which showed that both the overexpression of PTEN, a negative regulator of survivin, and survivin knockdown simultaneously inhibit the migration of FLS and IL-6 production (D. Chen et al, 2017). The STAT3mediated survivin expression in the inflammatory condition is not limited to the IL-6, other cytokines, including IL-11 and IL-22, can also induce survivin production through PI3K/Akt pathway and STAT3 activation (Nguyen, Putoczki, & Ernst, 2015).…”

“…The blocking of the following signal transducer and activator of transcription 3 (STAT3) signaling pathway leading to the induction of apoptosis and consequently, the level of survivin expression decreases (Aoki, Feldman, & Tosato, 2003). The regulation network of survivin is involved in the phosphoinositide 3-kinase (PI3K) and Akt pathways which is illustrated in Figure 2 (Chen et al, 2017). Suppression of RA-FLS-mediated by PTEN gene is also participated in survivin silencing (Chen et al, 2017) (Jackson & Bartek, 2009).…”

Suppression of RA-FLS-mediated by PTEN gene is also participated in survivin silencing (Chen et al, 2017) (Jackson & Bartek, 2009). Researchers indicated that DNA damage sensors play a key role in the recognition of foreign DNA aberrantly located in the cytoplasm to activate immune signaling.…”
