Rice fruit development is associated with an increased IAA content in pollinated ovaries

Abstract: To analyze fruit set and early fruit (caryopsis) development in rice, we established an in vitro spikelet culture system. The ovary of cultured pollinated spikelets grew rapidly and developed into fruits with an embryo and endosperm. When unpollinated spikelets were cultured on a medium containing 2,4-dichlorophenoxyacetic acid, parthenocarpic fruits lacking an embryo and endosperm developed. The number and size of the cells in the pericarp of parthenocarpic fruits were almost identical to those of fruits indu… Show more

“…IAA content is increased (approximately fivefold) in the rice ovary following pollination and the nucellus cells of unpollinated spikelets degenerate when cultured on medium containing auxin (Uchiumi and Okamoto, 2010), implying the role of IAA in nucellus degeneration. Considering that MADS29 expression is simultaneously upregulated ( Figure 8B) and the transcripts of IAA-responsive genes are reduced under MADS29 suppression ( Figure 8A, Table 2), it is supposed that MADS29 was induced by IAA after pollination to regulate the PCD of nucellus and nucellar projection and, hence, endosperm development ( Figure 8C).…”

“…The lemma, palea, lodicules, and filaments were removed and soaked in sterilized water for 2 min and then cultured in N6 medium containing IAA (Sigma-Aldrich) or 2,4-D (PhytoTechnology Laboratories) according to previous description (Uchiumi and Okamoto, 2010). The spikelets were cultured in greenhouse at 288C for 24 h under a 12-h-light/12-h-dark cycle.…”

“…In addition, fertilization drives the fruit set and is followed by fruit growth. 20 The following effects of fertilization are the increased synthesis of auxin and gibberellin in ovaries. The absence of fertilization (cut-male flowers) in this study resulted in a shortening of cob length.…”

“…Thus, both 2A12-iaaM gene expression and IAA content were increased in fruits, especially at fruit set. In many plant species, fruit set can be improved by auxin manipulation, either chemically or genetically (Molesini et al 2009;Uchiumi and Okamoto 2010). In this regard, the seedless fruits observed in Fig.…”
