RNA silencing is a homology-dependent gene inactivation mechanism that regulates a wide range of biological processes including antiviral defense. To deal with host antiviral responses viruses evolved mechanisms to avoid or counteract this, most notably through expression of viral suppressors of RNA silencing. Besides working as silencing suppressors, these proteins may also fulfill other functions during infection. In many cases the interplay between the suppressor function and other "unrelated" functions remains elusive. We will present host factors implicated in antiviral pathways and summarize the current status of knowledge about the diverse viral suppressors' strategies acting at various steps of antiviral silencing in plants. Besides, we will consider the multi-functionality of these versatile proteins and related biochemical processes in which they may be involved in fine-tuning the plant-virus interaction. Finally, we will present the current applications and discuss perspectives of the use of these proteins in molecular biology and biotechnology.

BackgroundRice black-streaked dwarf virus (RBSDV), a member of the genus Fijivirus within the family Reoviridae, causes severe damage to cereal crops in South East Asia. The protein P7-2, encoded by the second open reading frame of segment S7, is conserved among most plant-infecting fijiviruses, but its function is still obscure.ResultsIn this study, P7-2 was used as bait in two-hybrid screens of a cDNA library expressing Zea mays proteins. It was found that there is a strong interaction between P7-2 and Z. mays SKP1 (SKP1Maize), a core subunit of the multicomponent SCF (SKP1/Cullin1/F-box/Rbx1) E3 ubiquitin ligase. The interaction was then confirmed in leaf epidermal cells of Nicotiana benthamiana by bimolecular fluorescence complementation assay. Further investigations indicated that P7-2 also interacts with SKP1 proteins from other plants, including Arabidopsis thaliana, N. benthamiana,Oryza sativa and Saccharum sinense. The C-terminal fragment of SKP1Maize (residues 97–176) and the middle fragment of P7-2 (residues 79–214) are necessary to sustain the interaction, while the C-terminal putative α-helix domain spanning residues 214–295 of P7-2 greatly facilitates the interaction. Agrobacterium-mediated transient suppression assay showed that P7-2 has no obvious activity to suppress local RNA silencing.ConclusionsTaken together, our results indicated that RBSDV P7-2 can interact with SKP1 proteins from different plants. This is the first report linking a Fijivirus protein to a component of the ubiquitin proteasome system. P7-2 might be a potential F-box protein encoded by RBSDV and involved in the plant-virus interaction through ubiquitination pathway.

Virus-induced gene silencing (VIGS) is an attractive reverse-genetics tool for studies of plant gene function; however, efficient VIGS has only been accomplished in a few plant species. In order to extend the application of VIGS, we examined whether the use of a VIGS vector based on the tobacco rattle virus (TRV) would result in phenotypic changes in strawberry (Fragaria 9 ananassa). In this study we successfully silenced the expression of a strawberry phytoene desaturase (PDS) gene (FaPDS) and showed a significant reduction in FaPDS mRNA levels correlated with a concomitant photobleached phenotype. The spread of the virus harboring the TRV vector containing the coding sequence of the green fluorescent protein (GFP) marker protein (TRV-GFP) virus was readily monitored using fluorescent microscopy and a hand-held UV lamp, such that the infiltrated plants could be screened by detecting the GFP fluorescence. We also demonstrated that post-agroinfiltration water culture is an effective means to enhance the viability of vacuum-infiltrated plants. Taken together, the results confirm that VIGS using the TRV/TRV-GFP vector represents a new potentially valuable tool for strawberry functional genomic studies.
