Role of Actin Cortex in the Subplasmalemmal Transport of Secretory Granules in PC-12 Cells

Abstract: In neuroendocrine PC-12 cells, evanescent-field fluorescence microscopy was used to track motions of green fluorescent protein (GFP)-labeled actin or GFP-labeled secretory granules in a thin layer of cytoplasm where cells adhered to glass. The layer contained abundant filamentous actin (F-actin) locally condensed into stress fibers. More than 90% of the granules imaged lay within the F-actin layer. One-third of the granules did not move detectably, while two-thirds moved randomly; the average diffusion coeffic… Show more

“…The second possibility is that dynamin-induced actin reorganization retards granule recruitment at a later stage of secretion. This is supported by the positive role of actin in vesicle transport (64)(65)(66)(67) and the pronounced defect in the second-phase secretion (Figure 3), which largely relies on new granule recruitment to the PM (8,11). To directly test this idea, we incubated β cells for 1 hour with 20 mM glucose and 10 nM glucagon-like peptide-1 (GLP-1), an incretin peptide that increases insulin secretion through cAMPand VAMP8-dependent mechanisms (68).…”

“…The values of N f are typically N/3 (N f ¼ 16) (74), N/2 (N f ¼ 28) (75), and 2N/3 (N f ¼ 50), where N is the number of total MSD data points (34). As a precise measure of the dynamic parameters requires a sufficient number of MSD points, we choose N f ¼ 40 (N/2, N ¼ 80 (w ¼ 1.6 s and Dt ¼ 20 ms)) in calculating the dynamics parameters of directed and confined diffusion.…”

“…For example, in the neuronal PC12 cell line, myosin V was proposed to take over exocytic granules from microtubules and to ensure their anchoring 101 and short-range random movements 102 in the actin-rich cell cortex, thereby increasing the probability of membrane docking (FIG. 2).…”
