Role of HSP27 in tumor necrosis factor-α-stimulated interleukin-6 synthesis in osteoblasts

Abstract: Abstract. Heat shock protein 27 (HSP27) is known to act as a molecular chaperone. We have recently reported that HSP27 regulates osteocalcin synthesis in osteoblast-like MC3T3-E1 cells. In the present study, we investigated the role of HSP27 in tumor necrosis factor-α (TNF-α)-stimulated interleukin-6 (IL-6) synthesis in MC3T3-E1 cells. The levels of IL-6 release and IL-6 mRNA stimulated by TNF-α in MC3T3-E1 cells transfected with HSP27 was significantly higher than those in the control cells. In addition, the … Show more

“…Phosphorylated HSP27 was shown to participate in TNF-α-induced IL-6 synthesis in osteoblast-like MC3T3-E1 cells [111]. Overexpression of the phosphomimetic HSP27-3D mutant (in which serine residues 15, 78 and 82 are replaced by aspartic acid) resulted in higher IL-6 mRNA and protein levels than in cells overexpressing the non-phosphorylatable HSP27-3A mutant (in which serine residues 15, 78 and 82 are replaced by alanine).…”

“…The 27 kDa heat shock protein (Hsp27) is expressed in a differentiation-related pattern and can regulate osteocalcin synthesis ( 21 ). Activating transcription factor 4 (Atf4) is an important transcription factor that serves a pivotal role in osteoblastic differentiation and bone formation ( 22 , 23 ).…”
