Role of innate lymphoid cells and dendritic cells in intradermal immunization of the enterovirus antigen

Abstract: Enterovirus type 71 (EV71) and coxsackievirus A 16 (CA16) are the major pathogens of human hand, foot, and mouth disease (HFMD). In our previous study, intramuscular immunization with the inactivated EV71 vaccine elicited effective immunity, while immunization with the inactivated CA16 vaccine did not. In this report, we focused on innate immune responses elicited by inactivated EV71 and CA16 antigens administered intradermally or intramuscularly. The distributions of the EV71 and CA16 antigens administered in… Show more

“…The small pieces of skin were incubated in a digestion solution containing 5 mg/mL collagenase I (Sigma-Aldrich, St. Louis, MO, USA), 2.5 mg/mL trypsin (Thermo Fisher Scientific, Waltham, MA, USA), and 1 U/mL DNase I (Sigma-Aldrich) in Roswell Park Memorial Institute (RPMI) 1640 medium for 1.5 h at 37 °C with shaking at 200 rpm. The digested supernatant was filtered through a 70-μm cell strainer to obtain a single-cell suspension [ 15 ]. Furthermore, after removing both ends of the femur and tibia from the euthanized mice, the bone marrow was repeatedly flushed out into a Petri dish with PBS via a syringe until the bone was completely white.…”

“…[ 61 ] Unlike IPV, bOPV also provides intestinal mucosal immunity which is responsible for heterologous lymphocyte activation as gut epithelium has a reservoir of innate lymphoid cells which mimic the function of T-helper 2 cells. [ 64 ]…”

“…However, the interaction between other types of ILCs (ILC2s and ILC3s) with adaptive immune cells during viral infections also plays a crucial role during viral infections. The coordination between DCs and ILCs following intradermal injection with enterovirus type 71 (EV71) and coxsackievirus A 16 (CA16) Ags induces the successful adaptive immune response (neutralizing Abs and specific T cell response induction) as compared to the intramuscular injection 220 . EV71 and CA16 both are pathogens of the human hand, foot, and mouth disease (HFMD).…”
