Role of lung macrophages on susceptibility to respiratory infection following short-term moderate exercise training

Abstract: Moderate exercise training is associated with a decreased risk for upper respiratory tract infection in human and animal studies, but the mechanisms have not been elucidated. Lung macrophages play an important role in resistance to respiratory infection, and moderate exercise can enhance macrophage antiviral resistance, but no studies have directly tested the role of lung macrophages in this response. This study tested the effect of lung macrophage depletion on susceptibility to infection following short-term … Show more

“…Moreover, clodronate could have other unknown pharmacological effects that can affect compliance through mechanisms not involving macrophages. However, the use of clodronate is an effective and well-accepted technique for macrophage depletion and has been used successfully in many models (19,35,39). Also, we cannot completely eliminate the possibility that the clodronate may have been contaminated with endotoxin and thus participate in a preconditioning effect that could be protective against I/R.…”

“…This technique has not yet been used in studies involving exercise stress or any form of ␤-glucan. However, using this technique, we have recently shown an important role of macrophages on the beneficial effects of moderate exercise on susceptibility to infection (25).…”

“…Altered susceptibility to infection will be measured as changes in morbidity (time to sickness), symptom severity, and mortality (time to death). This was done using a murine model of exercise, respiratory infection (3,4,10,12), and in vivo lung macrophage depletion (25). The exercise protocol consisted of three consecutive days of prolonged treadmill running to fatigue designed to mimic a short period of severe exercise training.…”

“…5 The moderate intensity protocol was based on the protocols used in rats by other groups. 26,27 These protocols are well established in the literature, and their effects at the end of physical training has been previously described. 26,27 Considering respiratory mechanical function, we observed a significant decrease in Ers when comparing papain to saline groups.…”

However, we did not find significant differences between papain groups that were submitted to different intensities of exercise.…”
