Role of MYC in B Cell Lymphomagenesis

Abstract: B cell lymphomas mainly arise from different developmental stages of B cells in germinal centers of secondary lymphoid tissue. There are a number of signaling pathways that affect the initiation and development of B cell lymphomagenesis. The functions of several key proteins that represent branching points of signaling networks are changed because of their aberrant expression, degradation, and/or accumulation, and those events determine the fate of the affected B cells. One of the most influential transcriptio… Show more

“…Proto-oncoprotein c-Myc (hereafter Myc) is a master nuclear factor associated with cell proliferation, survival, and apoptosis [ 1 ]. Overexpression of Myc features numerous malignancies that include the highly aggressive B-cell lymphomas, such as Burkitt’s lymphomas, and also a subset of diffuse large B-cell lymphomas (DLBCL) [ 2 , 3 ]. Myc plays a central role in a large variety of malignancies, and therefore strategies aimed at targeting Myc have emerged as attractive cancer treatments.…”

“…In BL, high level of MYC and founder alterations induce a field defect characterized by the aberrant activation of PI3K signaling pathway and other changes mediated by the non-canonical binding of MYC to other proteins and genetic loci [ 29 , 53 , 54 ]. A constitutive and disregulated early expression of MYC and the synergism between MYC and the MYC-related miRNAs can overcome the transcriptional program mediated by BCL6.…”

“…MYC is re-expressed after (antigen) activation of the BCR, promoting proliferation and inhibiting differentiation of mature B cells [ 39 , 40 ]. After positive selection, MYC expression is downregulated, allowing B cells to differentiate into memory B cells or plasmablasts and to exit the germinal center [ 39 , 41 ]. In plasmablasts, MYC expression is suppressed, allowing terminal B cell differentiation into plasma cells [ 42 ].…”
