Role of serotonin 5-HT2A receptors in the development of cardiac hypertrophy in response to aortic constriction in mice

O. Lairez,
T. Cognet,
S. Schaak
et al.

Abstract: Serotonin, in addition to its fundamental role as a neurotransmitter, plays a critical role in the cardiovascular system, where it is thought to be involved in the development of cardiac hypertrophy and failure. Indeed, we recently found that mice with deletion of monoamine oxidase A had enhanced levels of blood and cardiac 5-HT, which contributed to exacerbation of hypertrophy in a model of experimental pressure overload. 5-HT2A receptors are expressed in the heart and mediate a hypertrophic response to 5-HT … Show more

“…41 Lairez et al demonstrated overexpression of the 5-HT 2A receptor in cardiomyocytes was transient and occurred during the early phase of hypertrophy in a model of transverse aortic constriction. 43 However, no differences were observed in baseline echocardiograms with regards to intraventricular septum dimensions or left posterior wall thickness. In addition, 5-HT 2B receptor blockade has been shown to reduce cardiac hypertrophy 44 and we demonstrated decreased mRNA levels of the 5-HT 2B receptor.…”

“…Our results show that serotonin leads to reduced CamKII activity, explaining the interference with clock functionality and hence with the differentiation process. This effect of serotonin on CamKII in BAT is novel; as in the heart, serotonin increases CamKII activity . However, the activation of CamKII was shown to be crucial for the differentiation of adipocytes, and its decrease as a result of serotonin treatment emphasizes the inhibitory role of serotonin in the differentiation process .…”

“…Moreover, it is important to point out that, in general, it is difficult to find a robust association and impact of just one SNP on elderly survival. However, in the case of the T102C‐SNP, previous studies performed in different populations indicate alterations in the concentration of HT‐2A receptors and association with chronic diseases and age‐related dysfunctions, that could affect the lifespan (Choi et al, ; Coto et al, ; Fehér et al, ; Jakubczyk et al, ; Lairez et al, ; Noronha et al, ; Prado‐Lima et al, ; Schwanke et al, ; Yamada et al, ).…”

“…In the vascular system, serotonin induces proliferation and migration via 5‐HT2A receptors activation and causes both enhancement of prostaglandin I2 production and reduction of nitric oxide (NO) by suppressing inducible NO syntheses (Machida et al, ). In a recent study, Lairez, Cognet, & Schaak, () showed that a selective blockade of 5‐HT2A receptors prevented the development of cardiac hypertrophy in mice. Another investigation by Nelson, Harrod, & Lamping, () suggested that both 5HT‐2A and 5HT‐2B receptors contribute to serotonin‐induced vascular dysfunction in diabetes.…”
