Role of serotonin type 1A receptors in fluvoxamine-induced inhibition of marble-burying behavior in mice

Abstract: This study examined the roles of presynaptic and postsynaptic serotonin type 1A receptors in fluvoxamine-induced inhibition of marble-burying behavior in mice. The effect of fluvoxamine was attenuated by the serotonin type 1A receptor antagonist WAY100635 at 1 mg/kg, while it was enhanced by the antagonist at 0.1 mg/kg. Fluvoxamine (30 mg/kg) and WAY100635 (0.1 and 1 mg/kg) did not affect spontaneous locomotor activity. These results suggest that the effect of fluvoxamine is mediated by postsynaptic serotonin … Show more

“…The number of marbles that have been buried is then counted by an observer, who is often blind to the treatment status of the subjects (Angoa-Pérez et al, 2013;Egashira et al, 2018;Millan et al, 2002). Importantly, since the majority of investigations do not distinguish between the specific behavioral patterns resulting in marbles being lowered into the burying substrate Gaikwad & Parle, 2011;Gomes, Casarotto, Resstel, & Guimarães, 2011;Harasawa, Ago, Itoh, Baba, & Matsuda, 2006;Honda, Kawaura, Soeda, Shirasaki, & Takahama, 2011;Li et al, 2006;Matsushima, Shirota, Kikura-Hanajiri, Goda, & Eguchi, 2009;Shimazaki, Iijima, & Chaki, 2004;Umathe et al, 2012;Yamada et al, 2002), the term Bburied^is generally applied to indicate the number of covered marbles. Although the reported methodologies always refer to the duration of exposure as well as the number of marbles and the burying substrate used, several experimental variables are subject to modification, either intentionally or not.…”

“…The pharmacological profile of mirtazapine is characterized by a potent antago- nism of presynaptic α 2 -adrenergic receptors on both noradrenaline and 5-HT neurons and by a potent antagonism of postsynaptic 5-HT 2 and 5-HT 3 but not 5-HT 1A receptors. It has been reported that the effect of fluvoxamine on marble-burying behavior is attenuated by the selective 5-HT 1A receptor antagonist WAY100635 at 1 mg/kg, while it is enhanced by WAY100635 at 0.1 mg/kg (13), suggesting that the involvement of 5-HT 1A receptors. Therefore, the effect of mirtazapine might be mediated, in part, by 5-HT 1A receptors.…”

“…Indeed, SSRIs such as fluvoxamine and paroxetine, which are used to treat human OCD symptoms (3), inhibit marble-burying behavior without affecting locomotor activity in mice (12,13). Moreover, uncompetitive NMDA-receptor antagonists such as memantine and amantadine inhibit marble-burying behavior without affecting locomotor activity (14).…”
