Role of tissue growth factors in aqueous humor homeostasis

Abstract: The aqueous humor supplies nutrients to the nonvascularized cornea, lens, and trabecular meshwork. A number of tissue growth factors have been detected in this fluid. The composition of these proteins changes dramatically with different ocular conditions, such as inflammation and glaucoma. In this review, an overview of new findings regarding effects of aqueous humor growth factors is given. Our main emphasis is on the regulation of the avascular anterior eye compartment, the possible role of growth factors in… Show more

“…It is important for the maintenance of both physiological function and metabolic homeostasis of anterior chamber structures including the corneal endothelium, trabecular meshwork, iris, and lens. 2 Previous studies have described alterations in the cytokine and chemokine compositions of the aqueous humor in various ocular diseases, such as uveitis, 3 branch retinal vein occlusion, 4 and age-related macular degeneration. 5 Increased levels of tumor necrosis factor (TNF)-a, 6 transforming growth factor (TGF)-b2, 7 interleukin (IL)-6, 8 IL-8, 8,9 serum amyloid A (SAA), 10 and vascular endothelial growth factor (VEGF) 11 have also been reported in glaucomatous aqueous humor.…”

“…8,9 Growth factors such as PDGF, FGF, and TGF-b are important for the growth of fibroblasts and blood vessels. 7 When macrophages encounter large foreign bodies, they may fuse to form a large multinucleated cell called the FBGC, the primary cellular mediators of the foreign body response.…”

“…22,23 There has been increasing interest in the role of proteins, including transforming growth factor beta (TGFb) and connective tissue growth factor (CTGF), in AqH homeostasis leading to raised IOP and glaucoma. 24, 25 Junglas et al 26 have used a transgenic mouse model to show that CTGF expressed in the AqH elevates IOP, which is associated with TM actin cytoskeleton modification. CTGF is a matricellular protein in that it interacts with and is induced by TGFb and it is through CTGF that TGFb mediates some of the downstream effects on proliferation, migration, and ECM production.…”
