Roles of Aluminum Hydroxide and Monophosphoryl Lipid A Adjuvants in Overcoming CD4+ T Cell Deficiency To Induce Isotype-Switched IgG Antibody Responses and Protection by T-Dependent Influenza Vaccine

Abstract: Vaccine adjuvant effects in CD4 deficient condition largely remain unknown. We investigated the roles of combined monophosphoryl lipid A (MPL) and Alum adjuvant (MPL+Alum) in inducing immunity after immunization of CD4-knockout (CD4KO) and wild-type (WT) mice with T-dependent influenza vaccine. MPL+Alum adjuvant mediated IgG isotype-switched antibodies, IgG secreting cell responses, and protection in CD4KO mice, which were comparable to those in WT mice. In contrast, Alum adjuvant effects were dependent on CD4… Show more

“…The levels of these innate acute inflammatory cells appeared to be lower than those reported in previous studies on adjuvants such as oilin-water emulsion MF59 and combination AS04, alum C monophosphoryl lipid A (MPL, a TLR4 ligand). 41 These intrinsic differences between F VLP and F protein in generating inflammatory microenvironment might be contributing to developing differential quality of immunity responsible for enhancing or preventing RSV vaccine-associated disease.…”

“…26 Both MF59 and AS04 adjuvanted influenza vaccinations were effective in inducing IgG isotype-switched antibodies and conferring protective immunity in a CD4 deficient mouse model. 13,27 MF59 was more potent than AS04 in exhibiting CD4independent adjuvant effects. AS04 showed a moderate level of CD4-dependency in inducing isotype-switched IgG antibodies, but AS04-adjuvanted T-dependent split influenza vaccine provided sufficient protection in CD4-deficient mice.…”
