RS1 Satellite Phage Promotes Diversity of Toxigenic Vibrio cholerae by Driving CTX Prophage Loss and Elimination of Lysogenic Immunity

Abstract: bIn El Tor biotype strains of toxigenic Vibrio cholerae, the CTX prophage often resides adjacent to a chromosomally integrated satellite phage genome, RS1, which produces RS1 particles by using CTX prophage-encoded morphogenesis proteins. RS1 encodes RstC, an antirepressor against the CTX repressor RstR, which cooperates with the host-encoded LexA protein to maintain CTX lysogeny. We found that superinfection of toxigenic El Tor strains with RS1, followed by inoculation of the transductants into the adult rabb… Show more

“…The deletion in the 2012Env-9/390 genomes encompasses CTXϕ and satellite phages TLCϕ and RS1ϕ, which are present in canonical toxigenic O1 strains18. The absence of CTXϕ core genes ( cep , orfU , ace , and zot ) and RS1 element ( rstR , rstA , rstB , and rstC ) were confirmed through BLAST searches of 2012Env-9/390.…”

“…All of the V. mimicus strains in this study were observed to produce replicative forms of both CTXФ ET and pre-CTXФ Env when induced by mitomycinC in the culture filtrates, which was detected by PCRs after DNAse and RNAse treatment (data not shown). On the other hand, like the El Tor strains of V. cholerae , the environmental V. mimicus strains also harbored rstC , i.e., the RS1 element, which has been recently observed to promote diversity by the loss of CTX prophage and lysogenic immunity to make room for new CTX prophage to be integrated (29). Presence of rstC has been shown to increase rstA transcription and CTXФ production (17), which may influence ctxAB transcription and diversification.…”

“…To our knowledge, the only other tyrosine recombinase for which efficient recombination between half-sites has been observed is Flp, the flippase of the 2-μm plasmid of Saccharomyces cerevisiae. Flp normally works as a homotetramer; however, in case of recombination between half-sites and full sites, the synapses imply three recombinases (27). In the case of Flp, trimer recombination is possibly related to the fact that active sites are assembled in trans (28).…”
