Rubidium-82 PET imaging is feasible in a rat myocardial infarction model

Abstract: Background. Small-animal myocardial infarct models are frequently used in the assessment of new cardioprotective strategies. A validated quantification of perfusion using a noncyclotron-dependent PET tracer would be of importance in monitoring response to therapy. We tested whether myocardial PET perfusion imaging is feasible with Rubidium-82 ( 82 Rb) in a small-animal scanner using a rat myocardial infarct model.Methods. 18 Sprague-Dawley rats underwent permanent coronary artery ligation (infarct group), and … Show more

“…Left ventricular ejection fraction (LVEF), left ventricular end-diastolic and end-systolic diameter (LVEDD and LVESD, respectively) were evaluated at a midpapillary short axis view. The range of post-ischemic hemodynamic parameters (e.g., LVEF) was similar to that found by others [39].…”

“…The myocardial infarctions were induced as described elsewhere [ 23 ]. In brief, the animals were anesthetized in 3–5% sevoflurane (AbbVie, Denmark) in an induction chamber before being intubated with a 16 G Venflon catheter (Vasofix® Safety, Braun, Denmark) with blunt needle and ventilated with 6–8 ml air/ventilation, at a frequency 80–90 ventilations/min (UNO microventilator-03, Netherlands).…”

“…To minimize 82 Rb activity from the blood pool, the PET list mode data was histogrammed into two time frames: the first 45 sec and the remaining 555 sec. Subsequently, the images were reconstructed as described previously [ 23 ].…”

“…Radionuclide tracers in rat myocardial infarcts such as cardiac 18F BMS747158-02 PET proved to estimate the PET defect size and left ventricular systolic and diastolic volume, that could provide a promising technique for the evaluation of MI in clinical trials ( 16 ). Using Rubidium-82 PET imaging in rats enables fast and time-efficient myocardial perfusion imaging after MI ( 17 ). Sequential evaluation of 18F-FDG PET and MRI in rat myocardial infarction previously showed to be a suitable approach to monitor cardiac remodeling in a multimodal way ( 18 ).…”
