Safe Mycobacterium tuberculosis DNA Extraction Method That Does Not Compromise Integrity

Abstract: Concern has been raised about the efficacy of the heat killing of mycobacteria during the isolation of DNA. We demonstrate a method that allows for the efficient killing of Mycobacterium tuberculosis without compromising DNA integrity for subsequent molecular investigation. This method reduces the risk of infection to the research scientist. The development of an internationally standardized method for the DNA fingerprinting of clinical isolates of Mycobacterium tuberculosis (6) has enabled scientists to gain i… Show more

“…Beijing genotype strains were subclassified as either “typical” or “atypical,” according to the presence or absence of an IS 6110 insertion in the noise transfer function (NTF) region ( 19 , 20 ). The atypical Beijing genotype strains were further classified by using the international standardized IS 6110 DNA fingerprinting method ( 21 ). For atypical Beijing strains that were drug-sensitive according to DST, sensitivity to isoniazid and rifampin was confirmed by sequencing the katG and rpoB genes.…”

“…M. tuberculosis isolates were cultured on MGIT (Becton Dickinson) or Löwenstein-Jensen medium, and DNA was extracted as previously described (32). Each isolate was classified by IS6110 RFLP genotyping (27) and spoligotyping (14) using internationally standardized protocols.…”
