Saikosaponin a Ameliorates LPS-Induced Acute Lung Injury in Mice

Abstract: The purpose of this study was to investigate the protective effects of Saikosaponin a (SSa), a triterpene saponin derived from Radix bupleuri, on lipopolysaccharide (LPS)-induced acute lung injury (ALI) using a murine model. The mice were given SSa 1 h after intranasal instillation of LPS. Then, lung histopathological examination, the wet/dry (W/D) ratio, myeloperoxidase (MPO), and inflammatory cytokines in bronchoalveolar lavage fluid (BALF) were detected in this study. The results showed that SSa reduced lun… Show more

“…Saikosaponins, an oleanane-type triterpenoid saponins, is the major bioactive ingredient extracted from Radix bupleuri , which has been used to prevent Alzheimer’s, pulmonary diseases, and even viral hepatitis [ 59 , 60 , 61 ]. However, in accordance with the cumulative evidence, saikosaponins probably contributed to toxicity in hepatocytes, and in particular, caused acute liver injury with doses of more than 19 g/day for a human being with 70 kg body weight.…”

“…Aside from that, the ethanol extract is more toxic than the aqueous extract [ 89 ]. A method of prolonged decoction was convincingly demonstrated to effectively detoxicate Polygonum multiflorum [ 59 ]. Based on these alterations in biomarkers, Polygonum multiflorum -induced disturbance in the metabolic process of fat, bile acid, and amino acid may be the dominant threats to the induction of oxidative hepatotoxicity.…”

“…The animal grouping strategy used in the current study was similar to a previous study (26). The dose of SSa administrated in the current study was determined according to a previous publication (16). SSa was administrated by intraperitoneal injection 1 h before inducing HP.…”

“…Emerging evidence has suggested that SSs display anti-inflammatory, antiviral, antioxidant, antitumor, immune regulatory and central nervous system protective effects (13,15). Among the SSs, SSa has attracted considerable attention due to its significant anti-inflammatory activity (16). Direct inhibition of proinflammatory cytokine expression and regulation of the inflammatory response via specific signaling pathways, such as the NF-κB signaling pathway, have been identified as critical mechanisms underlying the anti-inflammatory activity of SSa (17).…”

“…SSa also inhibits IL-1β-induced inflammatory responses in osteoarthritis chondrocytes by activating LXRα signaling (Gao et al, 2017), and inhibits LPS-induced production of reactive oxygen species (ROS), TNFα and IL-8 through the suppression of LPS-induced toll-like receptor 4 (TLR4) activation (Fu et al, 2015). In LPS-induced acute lung injury mice model, SSa reduced pathological injury via inhibiting TNFα and IL-1β and suppressing the NLR family pyrin domain containing 3 (NLRP3) inflammasome function (Du et al, 2017). Previous studies also demonstrated that SSa protects rats from carbon tetrachloride (CCl 4 )-induced liver injury and controls cortical impact-induced traumatic brain injury by suppressing hepatic inflammation and neural inflammation, respectively (Mao et al, 2016;Wu et al, 2010).…”
