Saponins as novel TNF-α inhibitors: isolation of saponins and a nor-pseudoguaianolide from Parthenium hysterophorus

Abstract: Two novel saponins and a 13-nor-pseudoguaianolide designated as hysterolactone were isolated from Parthenium hysterophorus. The two saponins were found to be potent inhibitors of TNF-alpha. Their mode of inhibition was studied through molecular modeling. The wet lab results were in concordance with the data obtained from docking experiments.

“…Some saponin rich extracts are well demonstrated to process anti-oxidative, anti-inflammatory (Wang et al, 2008) and anti-neuralgic (Kaur et al, 2010) effects in animal studies. Moreover, some saponins have also been reported to cause inhibition of calcium accumulation and reduction of TNF-a levels (Shah et al, 2009;Kaur et al, 2010). Further it has been well documented that massive rise in free radicals and calcium levels induce oxidative stress and neuro-inflammation (Honda et al, 2004;.…”

“…Results of analysis of three principal components between the levels of secondary metabolites (tannins (TAN), phenols (FEN), cyanogenic glycosides (GLUC), alkaloids (ALC) and saponins (SAP)) and cytotoxic activity expressed in LC 50 present in five selected medicinal plants. hysterophorus se reportó que fueron aisladas dos nuevas saponinas terpenoides, las cuales resultaron ser potentes inhibidores del factor de necrosis tumoral alfa (TNF-α) (Shah et al, 2009 (2003) no encontraron actividad hemolítica producida por el extracto de la planta. Para A. absinthium, C. aconitifolius y P. carpunya, solamente en la literatura se han reportado análisis cualitativos que indican la presencia de saponinas.…”

“…The decay in dynamic surface tension over increasing PHCE and PHNE concentrations indicates that both treatments were surface active and can reduce the surface tension of pure distilled water from 72.94 to 38.62 mN m 1 for PHCE and from 72.94 to 37.32 mN m 1 for PHNE. In this study, PHCE contained saponin 30 which work as a natural emulsifier by having hydrophobic parts from triterpene or steroid backbone covalently bond to hydrophilic parts such as carbohydrate groups 31 . While the present of surfactant Tensiofix and Tween 80 in PHNE have in-creased the ability of PHCE in reducing the interfacial tension of water.…”
