Schistosoma mansoni: histological analysis of the synergistic interaction between vaccine immunity and praziquantel therapy in the lungs of mice

Abstract: Naive CBA mice and mice vaccinated 4 weeks previously with gamma-irradiated cercariae of S. mansoni were challenged percutaneously with normal cercariae and then treated with 500 mg/kg body weight of Praziquantel (Pzq). The drug was administered intradermally on day 1 or intramuscularly on day 6, thus targeting against skin stage or lung stage challenge larvae respectively. The skin site of challenge and/or the lungs were removed at various time points to provide samples for histological examination. As report… Show more

“…Previous studies on the interaction between PZQ and the immune response have used an experimental model in which drug efficacy was enhanced by antisera or monoclonals administered concurrently with drug, the synergistically active immune effector mechanisms thus being putatively antibody (Doenhoff et al 1988, Brindley et al 1989). Enhancement of PZQ efficacy was also found in vaccinated mice (Flisser, Delgado & McLaren 1989, Piper, Mott & McLaren 1990). A model system involving passive transfer of antibodies, particularly heterologous antibodies as used in Table 1, has limitations.…”

“…Onion and garlic were administered in doses of 2g/100g body weight daily for 45 days in a standard pelleted diet containing 24% protein, 4% fat and about 4-5% fiber according to MANTAWY & MAHMOUD (2002). Praziquantel was administered in a dose of 500 mg/kg body weight on two successive days after 45 days of infection (PIPER et al, 1990).…”
