Understanding the mode of gene action that controls seed yield and Sclerotinia stem rot resistance in Indian mustard is critical for boosting yield potential. In a line × tester mating design, ten susceptible lines and four resistant testers were used to conduct genetic analysis. The significance of general combining ability (GCA) and specific combining ability (SCA) variances revealed that both additive and non-additive gene actions were involved in the inheritance of Sclerotinia stem rot resistance and yield attributing traits. In addition to 1000-seed weight and number of primary and secondary branches/plant, the genotypes RH 1569 (line) and DRMR 2035 (tester) appeared to be the strongest general combiners for Sclerotinia stem rot resistance. RH 1657 × EC 597317 was the only cross among several that demonstrated a significant desired SCA value for Sclerotinia rot resistance. Regarding SCA effects for yield and component traits, the cross RH 1658 × EC 597328 performed best, with a non-significant but acceptable negative SCA effect for resistance. DRMR 2035, RH 1222-28, RH 1569, RH 1599-41, RH 1657, RH 1658, and EC 597328 are promising genotypes to use as parents in future heterosis breeding and for obtaining populations with high yield potential and greater resistance to Sclerotinia stem rot disease in Indian mustard, based on GCA effects of parents, per se performance, and SCA effects of hybrids. Days to 50% flowering, number of primary branches/plant, main shoot length, and 1000-seed weight all had a high genotypic coefficient of variability (GCV), broad-sense heritability (h2bs), and genetic advance as percent of the mean (GAM) values, as well as significant and desirable correlations and direct effects on seed yield. As a result, these traits have been recognized as the most critical selection criterion for Indian mustard breeding programs.

Sclerotinia sclerotiorum provoca la podredumbre blanca del capítulo (PBC) en girasol, el cual le ofrece una resistencia de tipo cuantitativa. Consecuentemente, los programas de mejoramiento desarrollaran híbridos moderadamente resistentes a PBC para minimizar los daños potenciales de la enfermedad. La selección de híbridos se efectúa en ensayos multiambientales; no obstante, esta estrategia acarrea un gasto elevado. El objetivo fue hallar la asignación más adecuada de los recursos disponibles, aplicados en los ensayos tendientes a diferenciar híbridos por su nivel de resistencia a la PBC. Por ello en Balcarce se realizaron, durante 3 años, experimentos en el campo bajo un diseño en bloques completos aleatorizados con 3 repeticiones, en los que 37 híbridos fueron inoculados de forma asistida con S. sclerotiorum. A los capítulos enfermos se les midió el período de incubación relativo-PIR y el crecimiento diario relativo de la lesión-CLR (dos componentes de la resistencia parcial a aquella enfermedad). Se estimaron los BLUPs para cada híbrido, empleando todos los recursos disponibles (i.e. 108 plantas/híbrido, resultantes de la combinación 3 años, 3 repeticiones y 12 plantas/parcela), así como para aquellas combinaciones involucrando niveles menores de recursos. La prueba de la diferencia mínima significativa distinguió híbridos según sus BLUPs. Para ambas variables, hubo comportamientos diferenciales con la totalidad y con niveles menores de recursos. Los resultados mostraron la posibilidad de diferenciar, en un mismo ensayo, híbridos por su PIR y CLR ahorrando el 50% de los recursos disponibles al inicio de la experimentación, sin que la evaluación genotípica sea significativamente afectada.

Lily is a popular flower worldwide due to its elegant appearance and pleasant fragrance. Floral volatiles of lily are predominated by monoterpenes and benzenoids. While a number of genes for monoterpene biosynthesis have been characterized, the molecular mechanism underlying floral benzenoid formation in lily remains unclear. Here, we report on the identification and characterization of a novel BAHD acyltransferase gene that contributes to the biosynthesis of two related floral scent benzoate esters, ethyl benzoate and methyl benzoate, in the scented Lilium oriental hybrid ‘Siberia’. The emission of both methyl benzoate and ethyl benzoate in L. ‘Siberia’ was found to be tepal-specific, floral development-regulated and rhythmic. Through transcriptome profiling and bioinformatic analysis, a BAHD acyltransferase gene designated LoAAT1 was identified as the top candidate gene for the production of ethyl benzoate. In vitro enzyme assays and substrate feeding assays provide substantial evidence that LoAAT1 is responsible for the biosynthesis of ethyl benzoate. It was interesting to note that in in vitro enzyme assay, LoAAT1 can also catalyze the formation of methyl benzoate, which is typically formed by the action of benzoic acid methyltransferase (BAMT). The lack of an expressed putative BAMT gene in the flower transcriptome of L. ‘Siberia’, together with biochemical and expression evidence, led us to conclude that LoAAT1 is also responsible for, or at least contributes to, the biosynthesis of the floral scent compound methyl benzoate. This is the first report that a member of the plant BAHD acyltransferase family contributes to the production of both ethyl benzoate and methyl benzoate, presenting a new mechanism for the biosynthesis of benzoate esters.
