NOX1/NADPH oxidase regulates the expression of multidrug resistance‐associated protein 1 and maintains intracellular glutathione levels

Abstract: The involvement of superoxide‐generating NADPH oxidase (NOX) in the cytotoxic effects of cigarette smoke extracts has been documented. However, the underlying molecular mechanisms and NOX isoform involved have not been fully clarified. Among the different NADPH oxidase isoforms identified so far, NOX1 and NOX4 were found to be expressed in rat H9c2 cardiomyocytes. When H9c2 cells were exposed to acrolein or methyl vinyl ketone (MVK), major toxic components of cigarette smoke extracts, a dose‐dependent decline … Show more

“…Cytoplasmic CAT maintains the optimal metabolic balance by inhibiting superfluous H 2 O 2 and degrading hepatic fatty acids that reduce fat accumulation without oxidative damage (Vuolo et al., 2019). NADPH is a reducing agent in biosynthesis as a hydrogen transmitter and is the coenzyme of GSH reductase, maintaining the content of reducing GSH in cells (Wen et al., 2019). The content of NADPH is positively related to the content of reduced GSH fluctuates, and the dropped level of NADPH increases levels of ROS leading to tissue oxidative injury.…”

“…40 Similarly, neutrophils of CGD (a syndrome characterized by severe and repeated staphylococcal, coliform bacterial, and fungal infections) patients fail to kill microbes despite normal phagocytosis due to the defective NOX activation and inadequate intracellular GSH levels. 41,84 These studies suggest that maintaining redox balance in neutrophils by GSH system is critical while battling infection per se or chronic inflammatory conditions. Besides, GR plays a central role in the regeneration of GSH levels by reducing GSSG, thus maintaining the intracellular GSH pool and redox balance.…”
