Screening and evaluation of the strong endogenous promoters in Pichia pastoris

Abstract: Background Due to its ability to perform fast and high-density fermentation, Pichia pastoris is not only used as an excellent host for heterologous protein expression but also exhibits good potential for efficient biosynthesis of small-molecule compounds. However, basic research on P. pastoris lags far behind Saccharomyces cerevisiae, resulting in a lack of available biological elements. Especially, fewer strong endogenous promoter elements available for foreign protein expression or constructi… Show more

“…For instance, transcriptomics was used to screen strong endogenous promoter in Pichia pastoris. Based on transcriptomics analysis of Pichia pastoris cultured in three frequently-used media, ten potential strong endogenous promoters contributing to higher transcriptional expression levels were screened out [ 34 ]. In another example, due to limited constitutive promoters in Streptomycetes , transcriptomics was employed to screen more available constitutive promoters.…”

“…If a protein of interest is desired to be secreted, it is only required to add a secretion signal peptide at the N-terminus of the nascent polypeptide [24]. However, selecting an appropriate signal peptide is crucial since the protein quality and yield may vary widely depending on the heterologous protein being expressed [25]. Therefore, screening and characterizing many signal peptide sequences is a good approach to have adequate expression levels of different proteins.…”

“…Currently, there are three biosynthetic approaches that can be used to increase carotenoid production, including pathway engineering (metabolic engineering), host genetic modification, and protein engineering for enzyme and pathway optimization [103]. Metabolic engineering is the alteration of metabolic pathways with genetic modifications, such as the introduction of new genes with recombinant DNA technology.…”
