Screening of Potential Xanthine Oxidase Inhibitors in Gnaphalium hypoleucum DC. by Immobilized Metal Affinity Chromatography and Ultrafiltration-Ultra Performance Liquid Chromatography-Mass Spectrometry

Abstract: Abstract:In this study, a new method based on immobilized metal affinity chromatography (IMAC) combined with ultrafiltration-ultra performance liquid chromatography-mass spectrometry (UF-UPLC-MS) was developed for discovering ligands for xanthine oxidase (XO) in Gnaphalium hypoleucum DC., a folk medicine used in China for the treatment of gout. By IMAC, the high flavonoid content of G. hypoleucum could be determined rapidly and efficiently. UF-UPLC-MS was used to select the bound xanthine oxidase ligands in th… Show more

“…Interestingly, the complex had a more remarkable hypouricemic effect than luteolin and both compounds proved to be noncytotoxic to a human liver cancer cell line [70]. A glycosylated derivative, luteolin-4′-glucoside (Figure 18), also inhibited XO, with an in vitro IC 50 value of 0.26 μg/mL [71]. In addition, this molecule had potent uricosuric effects in hyperuricemic mice with renal mURAT1 and decreasing XO activity, as well as anti-inflammatory effects.…”

“…The ligands were then prepared for docking by computing the charges, and the structures were saved in PDBQT file format via AutoDock 4.2. Molecular docking experiment of ligands was performed following the protocol described by Zhang et al [ 25 ]. The size and center of the coordinates of the grid box was first validated by re-docking the co-crystallized ligand into the active sites of the receptor.…”

“…hypoleucum DC. is a folk medicine used for the treatment of gout in china and also used as anti-inflammatory, antitussive, gout and expectorant activities 12,18 . No comprehensive scientific study has been carried out on G. hypoleucum for their wound healing activity so far, hence the present study was undertaken to estimate the wound healing potential and compare their efficacy.…”
