Seeding of Human Vascular Cells onto Small Diameter Polyurethane Vascular Grafts

Abstract: Seeding of a mixed culture consisting of FBs and SMC resulted in an excellent EC adhesion and resistance to shear stress. Cell attachment was better on gamma-sterilized PUVPs compared to Eto-sterilization. The cells obviously maintained their ability to adapt to shear stress.

“…4 Recent procedures involved the pretreatment of prosthesis material with fibrin, collagen, or other matrix molecules that promote cell attachment and retention, but clinical relevance of these biomaterials have to be confirmed by large, human studies. 5,6 Endothelial progenitors alone or cocultured with myofibroblasts and fibrin preparations give also promising results. [7][8][9] Mesenchymal stem cells (MSCs) contribute to angiogenesis in the bone marrow and support hematopoietic cells in adult bone marrow.…”

“…The great importance of EC coverage of synthetic surfaces in tissue engineering has been shown in vitro [1821] as well as in animal studies [9,22]. One way to improve endothelialization of cell-repellent surfaces such as PU was nanoupgrading with titaniumcarboxonitride using PACVD technology [12,23].…”

“…Similarly importants for biocompatibility testing influence the sterilization process of the device with regard to the PUR surface properties 18, 19. The standard sterilization procedure for most pump chambers of artificial hearts is ethylene oxide.…”

“…Ethylene oxide is the standard sterilization procedure of PUR made pump chambers. Because Gulbins et al hypothesized that cell attachment was dependent on the sterilization procedure of PUR specimen,18, 19 we tested the cell attachment onto formaldehyde and ethylene oxide sterilized PUR. Our reference was PUR stored in sterilized H 2 O.…”
